摘要： This invention provides methods of quantitating nucleic acids from problematic samples, such as aged samples, formalin fixed samples, paraffin embedded samples, samples with aneuploid cells, and cells with fragmented nucleic acids. Methods include techniques to efficiently solublize the nucleic acids under non-denaturing conditions from preserved clinical samples without resort to organic extractions, to normalize cell counts regardless of aneuploidy, to access the fragmentation state of the nucleic acids, and to provide standard curves for degraded nucleic acid samples.

In one embodiment, an RNAscope® HPV assay was designed to detect the presence of E6/E7 mRNA of certain high-risk HPV subtypes related to head and neck cancer. The present invention also provides a method and a kit for determining whether a cervical lesion is a benign lesion or a cervical intraepithethial neoplasm lesion. In one embodiment, an RNAscope® HPV assay was designed to detect the presence of E6/E7 mRNA of certain high-risk HPV subtypes related to cervical cancer. The present invention further provides a method for determining the progression of cervical intraepithethial neoplasm based on the spatial pattern and levels of the E6/E7 mRNA of certain high-risk HPV subtypes. The present invention also provides a method for determining the risk of developing cervical cancer in a human diagnosed with cervical intraepithethial neoplasm based on presence and absence of the certain subgroups of high-risk HPV subtypes.

摘要： The invention provides methods and kits for detecting and/or quantifying nucleic acid sequences of interest by using pairs of probes containing donor-acceptor moieties that when hybridized on a target polynucleotide, with one of the probes being hybridized to a sequence of interest in the target polynucleotide, places the donor-acceptor moieties in sufficiently close proximity such that a detectable signal is generated. Methods of the invention are particularly useful for genotyping analysis and gene expression profiling. Methods of the invention are easily adaptable to arrays and automation. The invention also provides probes with enhanced hybridization efficiency and/or specificity comprising a probe portion having a spacer element and/or a minor groove binder molecule, and methods and kits for using these probes.

摘要： Disclosed are systems and methods involved in extreme high throughput screening of compounds which have an affinity for a biological target. The system is based on a capillary bundle with two distinguishable ends wherein capillaries on one end are connected to compounds stored in discrete reservoirs and capillaries on the other end are bound and processed to form a two dimensional microarray. A capillary bundle having reaction wells for hybridization and compound reaction in one end of the capillaries is disclosed. The capillaries may be light-condicting. Also disclosed are various methods of identifying a target compound in a liquid using this capillary bundle as well as methods of fabricating the bundle. A novel surface tension guided reaction chamber is also provided. Methods and chemistry for fabrication and use of a surface tension guided reaction chamber in binding and hybridization assays are also disclosed. Methods and systems for precise metering of fluids within the capillaries and at the reaction chambers, including the surface tension guided "virtual" reaction well is provided. Methods for performing high throughput screens using optical fiber lined capillaries of the invention are also provided. The capillary arrays are used with both liquid probes as well as probes immobilized on the walls of the capillary.

The system has a print-head (102, 606, 706, 806) formed of one or more capillary bundle (104), such as light-guiding capillaries. Methods of registering or correlating the distal 112 and proximal 110 ends of the capillaries are also provided.
