摘要： An apparatus for amplifying nucleic acids includes a substrate, a reaction vessel formed inside of the substrate, at least one first inlet channel formed inside the substrate, connected to an end of the reaction vessel, and allowing introduction of a reactant aqueous solution containing reactants for nucleic acid amplification into the reaction vessel, a second inlet channel formed inside the substrate, connected to the end of the reaction vessel, and allowing introduction of a fluid that is phase-separated from the reactant aqueous solution and does not participate in amplification reaction into the reaction vessel, and a heating unit installed on the substrate in such a way to thermally contact with the substrate and heating the substrate, wherein the reactant aqueous solution contacts the fluid to create a plurality of droplets surrounded by the fluid in the reaction vessel and the nucleic acids are amplified in the reactant aqueous solution droplets.

Interconnect input/output ports [12] of the fluidic interconnect backbone [10] are interfaced with microchannel input/output ports [14] of the fluidic microstructures at a plurality of non-polymeric interconnect seals [50]. Interconnect microchannels [15] are defined entirely by the fluidic interconnect backbone [10] and extend between the non-polymeric interconnect seals [50] without interruption by additional sealed interfaces. At least one of the fluidic microstractures [20, 30, 40] may comprise a mixing microstructure formed by a molding process. Another of the fluidic microstructures [20, 30, 40] may comprise an extruded reactor body. Still another fluidic microstructure [20, 30, 40] may comprise a quench-flow or hydrolysis microreactor formed by a hot-pressing method.

Controlling dispersion of reagents within fluid streams can include flowing multiple fluid streams each including reaction reagents into contact through a mixing region to laterally mix the fluid streams and then passing the merged, laterally mixed fluid stream through a controlled dispersion element to axially disperse the reaction reagents merged fluid stream. Controlling dispersion of reagents within fluid streams can include controlling flow rates of multiple fluid streams each including reaction reagents to create a concentration gradient that is substantially free of a measurable dispersion artifact. The biochemical reaction can occur in a microfluidic chip.
