摘要： Methods and compositions are provided for inhibiting, preventing and treating amyloid depositions, e.g. in pancreatic islets, wherein the amyloidotic deposits are islet amyloid polypeptide (IAPP)-associated amyloid deposition or deposits. Accordingly, the compositions and method of the invention are useful for inhibiting diabetes. The invention also provides a process for the preparation of cells suitable for transplantation into a mammal, which cells are capable of forming fibrils, said process comprising contacting the cells with an inhibitor of fibril formation. In particular the process prepares cells for use in a method of treating diabetes. Also provided are a culture medium comprising the inhibitor more particularly selected from the group consisting of 3-(3-hydroxy-1-propy)amino-1-propanesulfonic acid; DL-2-amino-5-phosphovaleric acid; 4-Phenyl-1-(3'-sulfopropyl)-1,2,3,6-tetrahydropyridine, cyclohexylsulfamic acid, O-phospho-L-serine, hexafluoroglutaric acid, 8-methoxyquinoline-5-sulfonic acid, 3-amino-2hydroxy-1-propanesulfonic acid, and 3-dimethylamino-1-propanesulfonic acid and 1,2,3,4-tetrahydroisoquinoleine.

摘要： Methods for identifying ATP-binding cassette A1 (ABCA1) agonists using pancreatic B-cells capable of expressing ABCA1 are disclosed. Methods for increasing the expression of ABCA 1 in pancreatic B cells by providing an expression vector comprising pancreatic B-cell control sequences and a nucleic acid encoding ABCA1 are also disclosed. Pancreatic B-cells which express ABCA1 and agonists of ABCA 1 which modulate pancreatic B-cell function are used in treating and preventing the onset of type 2 diabetes mellitus.

摘要： The invention features methods for the treatment of diabetes by administering a IAP nucleic acid sequence to a subject. One methodincludes the steps of. (a) providing one or more insulin-producing cells or one or more precursor cells capable of producing progeny cells tha tproduce insulin; (b) transducing the cells with a nucleic acid sequence encoding an IAP plypeptide, the nucleic acid sequence postioned for expression of the IAP polypeptide in the cells, wherein expression of the IAP polypeptide increases survival of the cells relative to untreated control cells; and (c)transplanting the cells from step (b) that have been transduced with anucleic acid sequence encoding an IAP polypeptide into the individual, wherein the cells produce insulin in amounts sufficient to treat diabetes in the individual.
