    • The present invention provides a biointerface membrane for use with an implantable device that interferes with the formation of a barrier cell layer and instead forms a foreign body response (FBR) layer including: an innermost FBR layer (40) composed generally of macrophages and foreign body giant cells (41) that forms over the entire surface (48a) of the membrane (48); an intermediate FBR layer (42) composed primarily of fibroblasts (43) and fibrous matrix (44); and an outermost FBR layer (46) is loose connective granular tissue containing new blood vessels (45); wherein the innermost layer (40) is resistant to cellular attachment and is impermeable to cells and wherein the outermost layer (46) supports tissue attachment and interferes with barrier cell layer formation. In addition, the present invention provides sensors including the biointerface membrane, implantable devices including these sensors or biointerface membranes, and methods of monitoring glucose levels in a host utilizing the analyte detection implantable device of the invention. Other implantable devices which include the biointerface membrane of the present invention, such as devices for cell transplantation, drug delivery devices, and electrical signal delivery or measuring devices are also provided.

    • The invention relates to a sensor for the in vivo measurement of an analyte, comprising a plurality of particles of suitable size such that when implanted in the body of a mammal the particles can be ingested by macrophages and transported away from the site of implantation, each particle containing the components of an assay having a readout which is an optical signal detectable transdermally by external optical means, and either each particles being contained within a biodegradable material preventing ingestion by the macrophages, or each particle being non-biodegradable. The invention relates to a process for the detection of an analyte using such a sensor, comprising implantation of the sensor into the skin of a mammal, transdermal detection of analyte using external optical means, degradation of the biodegradable material, ingestion of the particles by macrophages, and removal of the particles from the site of implantation by macrophages.

    • An electrochemical sensing system includes a working electrode and a reference electrode, which can at least partially be disposed in a housing. At least a portion of the working electrode includes rhodium metal. An electrical circuit is disposed in the housing and configured to be electronically coupled to the electrodes. The electrical circuit is operative to: (a) bias the working electrode at a voltage of less than about 0.4 V, and (b) measure a current corresponding to the concentration of the target analyte. A communications module is electrically coupled to the electrical circuit and configured to display a concentration of the target analyte, and/or communicate data between the electrical circuit and an external device. The electrodes are movable between a first configuration in which the electrodes are substantially disposed inside the housing, and a second configuration in which at least a portion of the electrodes is disposed outside the housing.
