摘要： An assay method and device can perform at least one (e.g., at least two) assays on a single aliquot of a sample liquid. The device can mix a sample liquid with assay reagents including magnetically susceptible particles. The device is configured to create a sample liquid-air interface with the sample liquid. The magnetically susceptible particles can be located (via an applied magnetic field) at the liquid-air interface when a second liquid contacts the interface to form a liquid-liquid interface. The magnetic particles travel across the liquid-liquid interface to the second liquid. The magnetically susceptible particles are configured to trans-port an analyte across the interface into the second liquid. An assay for the analyte is performed in the second liquid. An assay for another analyte can also be performed in the sample liquid.

摘要： The assay device (10) comprises a micro flow path (76); a porous medium provided near the distal end portion (80) of the micro flow path (76); and a space (82) provided between the micro flow path (76) and the porous medium for controlling the flow rate of a fluid moving from the micro flow path (76) to the space (82). After a fluid moved along the micro flow path (76) based on a lateral flow is brought into contact with the porous medium beyond the space (82) and is absorbed to the porous medium, the fluid is divided by the space (82) so that the fluid stays in the micro flow path (76). With this structure, it is possible to perform solution exchange in the micro flow path without using an external device such as a pump.

摘要： The invention provides a passive fluidics circuit for directing different fluids to a common volume, such as a reaction chamber or flow cell, without intermixing or cross contamination. The direction and rate of flow through junctions, nodes and passages of the fluidics circuit are controlled by the states of upstream valves (e.g. opened or closed), differential fluid pressures at circuit inlets or upstream reservoirs, flow path resistances, and the like. Free diffusion or leakage of fluids from unselected inlets into the common outlet or other inlets at junctions or nodes is prevented by the flow of the selected inlet fluid, a portion of which sweeps by the inlets of unselected fluids and exits the fluidics circuit by waste ports, thereby creating a barrier against undesired intermixing with the outlet flow through leakage or diffusion. The invention is particularly advantageous in apparatus for performing sensitive multistep reactions, such as pH-based DNA sequencing reactions.

摘要： A microfluidic device is provided for inducing the separation of constituent elements from a microfluidic sample by introducing phase changes in the microfluidic sample while contained in a microfluidic channel in the device. At least a portion of the microfluidic sample is frozen to cause fractional exclusion of the constituent element from the frozen portion of the microfluidic sample. Different portions of the microfluidic sample may be frozen in different sectors and at different times in order to cause movement in a desired direction of the separated constituent element. Portions of the microfluidic sample may be frozen in a sequential order of adjacent sectors within the microfluidic channel in order to cause sequential movement of the excluded constituent element toward one portion of the microfluidic channel. The frozen portion of the microfluidic sample is then thawed, wherein the separated constituent element remains substantially separated from the thawed, purified microfluidic sample.

摘要： Emulsion breaking and phase separation is achieved by droplet adhesion. An emulsion breaking device includes a channel having distinct adjacent zones with distinctly different surface wettability characteristics, namely, solvophilic and solvophobic surfaces. The device is positioned such that the upstream portion of the device is configured to be wetted by the continuous phase of the emulsion, and the downstream portion of the device is configured to be wetted by the dispersed phase of the emulsion. As the emulsion flows from the upstream zone to the downstream zone, the change in surface wettability characteristics promotes adhesion of the dispersed phase as the dispersed phase wets the surface of the downstream portion of the channel, which results in breaking of the emulsion. Subsequent collection of the broken emulsion in a collection vessel results in separation of the disparate phases to facilitate their recapture and recycling.
