    • Disclosed are devices and methods for administration of charged drugs to tissues of the oral cavity and for intra-tissue administration.The disclosed drug applicator for intra-tissue use includes an introducer such as a needle or catheter that is adapted to penetrate into a living tissue. The introducer has a drug-providing portion, and the applicator has current-generating element to pass through it an electrical current. Passing an electrical current through the drug-providing portion causes the drug to eject into treated tissue.Also disclosed are charged drugs suitable for iontophoretic administration, which include a bioactive agent encapsulated in charged nano-particles.

    • A polymeric carrier for delivery of bioactive or bioreactive molecules is provided, including a stereocomplex of one or more biocompatible polymers and having incorporated on or within the complex the molecules to be delivered. In a preferred embodiment, the biocompatible stereoselective polymers are linear or branched D-PLA homo- and block-polymers, linear or branched L-PLA homo- and block-polymers, copolymers thereof, or mixtures thereof, in stereocomplexed form. In one preferred embodiment, the polymeric carrier is complexed with a complementary stereospecific bioactive molecule. In other embodiments, the bioactive, or bioreactive (for example, for use in diagnostic applications), is bound to the complex by ionic, hydrogen, or other non-covalent binding reactions not involving stereocomplexation, or is physically entrapped within the complex, either at the time of complex formation or when the polymeric material is formulated into particles, tablets, or other form for pharmaceutical application. Exemplary bioactive molecules include peptides, proteins, nucleotides, oligonucleotides, sugars, carbohydrates, and other synthetic or natural organic molecules, as well as stereoselective drugs of a molecular weight of 300 daltons or higher. Examples demonstrate preparation of stereocomplexes, as well as their use for controlled and/or sustained release.

    • Branched polyanhydrides that have superior properties for use in the controlled delivery of substances, prepared by the polymerization of a dicarboxylic acid monomer and a branching agent. The branching agent is a polycarboxylic acid monomer such as 1,3,5-benzenetricarboxylic acid ("BTC"), or an oligomerized fatty acid trimer. Alternatively, the branching agent is a polycarboxylic acid polymer such as poly(acrylic acid) ("PAA"). These branched polymers have significantly higher molecular weights and lower specific viscosity than linear polymers prepared under similar conditions. A major advantage of branched polyanhydrides is that the degradation and release kinetics can be substantially altered without significantly changing the physical properties of the polymer relative to the corresponding linear polymer. By manipulating both the diacid and the branching agent, a wide variety of biodegradable controlled delivery devices for diverse applications can be prepared.
