    • PURPOSE: A primer for diagnosing ASSVd(apple scar skin viroid) infection and a diagnosis method using the same are provided to accurately detect infection by ASSVd and improve diagnosis accuracy. CONSTITUTION: A primer for diagnosing ASSVd(apple scar skin viroid) comprises a sequence number 1 or 2. A method of NASBA(nucleic acid sequence based amplification) for diagnosing infection by ASSVd comprises: a first step of annealing a first primer on a sense RNA and synthesizing single-stranded DNA molecule; a second step of reacting RNase to DNA-RNA hybrid molecule to remove RNA; a third step of annealing a second primer on single-stranded DNA molecule and reacting reverse transcriptase to synthesize double stranded DNA; and a step of synthesizing antisense RNA mediating RNA polymerase with single-stranded DNA molecule as an template.

    • PURPOSE: A development method of SCAR marker linked to a gene for columnar growth habit in apple is provided, which method allows for discrimination of the columnar growth habit in young plant of apple. CONSTITUTION: The development method of SCAR(sequence characterized amplified region) marker linked to a gene for columnar growth habit in apple comprises the steps of: cross-fertilization of a standard type apple of "Fuji" with a columnar growth type apple of "Tuscan" to obtain a descendant apple F1, and analyzing properties of the descendant apple F1; selecting a marker linked to a gene for columnar growth habit through RAPD(Random amplified polymorphic DNA) analysis using BSA(bovine serum albumin); and sequencing the cloned RAPD marker, wherein the RAPD marker linked to the gene for columnar growth habit is a nucleotide sequence WB82670 of SEQ ID NO:1; and the SCAR marker is a pair of SCAR primers having the nucleotide sequences of SEQ ID NO:2 and SEQ ID NO:3.
