    • PURPOSE:To measure the amount of a polyamine contained in blood serum, etc.,, with sensitivity of 3 times, by forming 2 molecules of reduced from nicotinamide adenine dinucleotide (NADH) and one molecule of reduced form nicotinamide adenine dinucleotide phosphate (NADPH) from one molecule of the polyamine. CONSTITUTION:Putrescine oxidase is reacted with a specimen, such as blood serum, containing a polyamine, to form hydrogen peroxide and an aminoalkylaldehyde. The hydrogen peroxide is then reacted with methanol, catalase, formaldehyde dehydrogenase and nicotinamide adenine dinucleotide (NAD ) to form NADH. On the other hand, the aminoalkylaldehyde is converted into NADH with a dehydrogenase and the formed aminoalkylcarboxylic acid is simultaneously reacted with aminoalkylcarboxylic transaminase and succinic semialdehyde dehydrogenase in the presence of alpha-ketoglutaric acid and nicotinamide adenine dinucleotide (phosphate) [NAD(P) ] to form NAD(P)H. The resultant 2 molecules of the NADH and one molecule of the NADPH are measured to determine the polyamine.
