摘要： PURPOSE:The titled standard composition, containing a milk protein, glycoprotein derived therefrom or enzymic hydrolyzate thereof prepared from a relatively inexpensive and readily available substance as a raw material, having stabile quality and useful for diagnosing inflammations, observing progress of symptoms, etc. CONSTITUTION:beta-Lactoglobulin separated from whey in cow's milk as a milk protein, glycoprotein derived therefrom or enzymic hydrolyzate thereof is dissolved in a tris buffer solution at 8.0pH and trypsin dissolved in deionized water is added and reacted at 35 deg.C for 3hr. The reaction mixture is then centrifuged to give a supernatant, which is then dialyzed with deionized water and freeze dried to afford the aimed standard substance containing bonded type sialic acid. Tris buffer solution (7.0pH), neuraminidase, N-acetylneuraminic acid aldolase and reduced form of nicotinamide adenine dinucleotide (NADH) are added and reacted with the standard substance as a test solution to measure absorbance thereof at 340nm wavelength.

摘要： PURPOSE:To simply and highly sensitively perform the quantitative determination of hydrogen peroxide, by allowing an alcohol and catalase to act on an H2O2-containing specimen and subsequently forcing luciferase to act on the resultant aldehyde in the presence of a long chain alkyl compound and FMNH2 to measure the amount of the emitted light. CONSTITUTION:Catalase is allowed to act on a specimen containing hydrogen peroxide in the presence of an alkylalcohol (e.g., methanol) and the luciferase of a luminous bacterium (e.g., VIbrio harveyi) is forced to act on the produced aldehyde in the presence of a long chain alkyl compound (e.g., decane) and FMNH2 (reduction type flavin monoucleotide). The resultant light is measured with an usual emitted light-measuring apparatus such as a lumiphotometer to determine the quantity of the hydrogen peroxide in the specimen.

摘要： PURPOSE:To measure the amount of a polyamine contained in blood serum, etc.,, with sensitivity of 3 times, by forming 2 molecules of reduced from nicotinamide adenine dinucleotide (NADH) and one molecule of reduced form nicotinamide adenine dinucleotide phosphate (NADPH) from one molecule of the polyamine. CONSTITUTION:Putrescine oxidase is reacted with a specimen, such as blood serum, containing a polyamine, to form hydrogen peroxide and an aminoalkylaldehyde. The hydrogen peroxide is then reacted with methanol, catalase, formaldehyde dehydrogenase and nicotinamide adenine dinucleotide (NAD ) to form NADH. On the other hand, the aminoalkylaldehyde is converted into NADH with a dehydrogenase and the formed aminoalkylcarboxylic acid is simultaneously reacted with aminoalkylcarboxylic transaminase and succinic semialdehyde dehydrogenase in the presence of alpha-ketoglutaric acid and nicotinamide adenine dinucleotide (phosphate) [NAD(P) ] to form NAD(P)H. The resultant 2 molecules of the NADH and one molecule of the NADPH are measured to determine the polyamine.

摘要： PURPOSE:To determine both a diamine and a polyamine by a simple device by a simple operation in a short time, by treating a test specimen with a di- and/or polyamine oxidase, treating formed H2O2 with catalase in the presence of an alcohol and measuring the formed aldehyde. CONSTITUTION:A test specimen is treated with a diamine oxidase (e.g., produced by germinated soybean or by organisms) and/or a polyamine oxidase (e.g., produced by animal source such as bovine plasma, etc.). Then formed H2O2 is treated with a catalase (e.g., preferably produced by animal source) in the presence of an alcohol such as methanol, ethanol, etc., the formed aldehyde is measured to determine a diamine and a polyamine. The measurement of the aldehyde is carried out by chemical method wherein direct colorimetry is used. The determination of this invention is applicable to an automatic analyzer, so it an be adopted widely to day-to-day, clinical diagnosis method and utilized for detecting, diagnosing and remedying cancers.
