摘要： PURPOSE: To obtain the titled compound important th respect to physiological activity and nutritive physiology industrially advantageously, by desalting a filtrate of ultrafiltration of a milky raw material substance containing an oligosaccharide of sialic acid bond by electrodialysis, treating it with an anion exchange resin, adjusting it to a proper pH, desalting it again by electrodialysis.
CONSTITUTION: A filtrate of ultrafiltration of milky raw material substance or molasses prepared in the process of lactose, containing an oligosaccharide of sialic acid bond, is desalted by electrodialysis, passed through a column packed with an anion exchange resin, so that the oligosaccharide of sialic acid is adsorbed on the resin. Water is then passed through the column to remove neutral saccharide. An eluting solution such as hydrochloric acid, etc. is passed through the column to elute the oligosaccharide. The eluate is adjusted to a pH approximately neutrality, and desalted agai by electrodialysis to give the desired oligosaccharide.

The reaction mixture is then centrifuged to give a supernatant, which is then dialyzed with deionized water and freeze dried to afford the aimed standard substance containing bonded type sialic acid. Tris buffer solution (7.0pH), neuraminidase, N-acetylneuraminic acid aldolase and reduced form of nicotinamide adenine dinucleotide (NADH) are added and reacted with the standard substance as a test solution to measure absorbance thereof at 340nm wavelength.
