摘要： NEW MATERIAL:The compound of formula I (Gal is galactose; Fruf is fructofuranose; n is 0 or 1) having the following characteristics. Molecular weight, 504 (n=0) or 666 (n=1); solubility, soluble in water and insoluble in benzene, acetone, etc.; color reaction, positive to phenol sulfuric acid method, etc., and negative to ninhydrin reaction, etc. EXAMPLE:beta-D-galactopyranosyl-(1 6)-beta-D-galactopyranosyl-(1 4)-D-fructofura noside of formula II. PREPARATION:The oligosaccharide of formula III (Glc is glucose) used as a starting raw material is made to react with an alkali such as NaOH preferably at 20-60 deg.C for 2-10 hr to effect isomerization of the material and the reaction mixture is neutralized. After converting the unreacted oligosaccharide in the reaction mixture to an aldonic acid with a weak oxidizing agent, the reaction mixture is desalted and subjected successively to ion exchange resin treatment and activated carbon column treatment to separate the objective compound.

摘要： PURPOSE:To separate and purify human lactoferrin in high yield and purity, from human milk, by using a specific affinity column. CONSTITUTION:A monoclonal anti-human lactoferrin antibody produced by a hydridoma is immobilized to an insoluble carrier [e.g. Affigel-10(Biolad Corp.), etc.], and is used as an affinity column to separate and purify human lactoferrin from human milk. The above antibody can be prepared by fusing the spleen cell of mouse immunized with human lactoferrin and a myeloma cell, administering the resultant hybridoma to mouse, and recovering and purifying the ascites of the mouse. EFFECT:The objective compound can be separated easily from the other milk components only by passing through a column once, without using pretreatment. USE:Preventive and remedy for the infective diseases of infants caused by iron- demanding pathogen.

The recovered fraction containing the lactoferrin and lactoperoxidase is subsequently combined with the fraction containing the immunoglobulin and desalting and removal of a low-molecular fraction are carried out by using an ultrafiltration membrane having >=50kD to recover a lactenin fraction containing the active immunoglobulin, lactoferrin and lactoperoxidase.
