    • PURPOSE:To determine saccharified protein in a sample simply, quickly and with high accuracy by a method wherein the sample which contains the saccharified protein and glucose coexisting with the saccharified protein is oxidation- treated, it is reacted with hydrazine, phenylhydrazine and a nonionic surface-active agent are added and a caused change in absorbance is measured. CONSTITUTION:A sample as blood which contains saccharified protein and which is originated from a living body, preferably blood serum, is prepared as a sample to be used. The sample is first oxidation-treated, and glucose which affects the measurement of the saccharified protein is removed. Hydrazine is reacted with the sample, and the saccharified protein in the sample is changed into free 2-ketoglucose. Then, phenylhydrazine is added, glucose diphenylhydrazone (a yellow pigment) is produced, and the precipitation of protein is prevented by a nonionic surface-active agent. The reaction rate of this coloration reaction is nearly definite for five to ten minutes from the start of the reaction, and a standard curve is linear. As a result, when a change in absorbance during this period is measured, the colorimetric determination of the saccharified protein in the sample can be performed with high accuracy.
