    • PROBLEM TO BE SOLVED: To provide an improved apparatus for implementing qualitative and quantitative analyses of a microscopic biological sample, and a method thereof. SOLUTION: The apparatus and method is for separating, immobilizing, and quantifying a biological substance from within a fluid medium. The biological substance is observed by employing a vessel 6 incorporating a chamber, the vessel including a transparent collection wall 5. A magnetic capture structure with a high internal gradient is on the transparent collection wall 5 and magnets 3 produce an externally-applied force for transporting a magnetically responsive material toward this transparent collection wall 5. This magnetic capture structure includes a conductor means supported on the transparent collection wall 5 in order to enable electrical manipulation of the biological substance. This chamber includes one compartment or a plurality of compartments with different heights. The chamber may have a porous wall.     • PROBLEM TO BE SOLVED: To provide a system for enumeration of cells in fluids by image cytometry for assessment of target populations such as leukocyte partial group in different bodily fluids, or bacterial contamination in environmental samples, food products and bodily fluids.
      SOLUTION: Fluorescently labeled target cells are linked to magnetic particles or beads. In one embodiment, a small permanent magnet is inserted directly into the chamber containing the labeled cells. The magnets are coated with PDMS silicone rubber to provide a smooth and even surface which allows imaging on a single focal plane. The magnet is removed from the sample and illuminated with fluorescent light emitted by the target cells captured by a CCD camera. In another embodiment, a floater having a permanent magnet makes the target cells array along a single imaging plane within the sample solution. An image analysis can be performed with a novel algorithm for measuring cells on the surface reflecting the target cell concentration of the original sample.
