摘要： PROBLEM TO BE SOLVED: To provide a system for enumeration of cells in fluids by image cytometry for assessment of target populations such as leukocyte partial group in different bodily fluids, or bacterial contamination in environmental samples, food products and bodily fluids.
SOLUTION: Fluorescently labeled target cells are linked to magnetic particles or beads. In one embodiment, a small permanent magnet is inserted directly into the chamber containing the labeled cells. The magnets are coated with PDMS silicone rubber to provide a smooth and even surface which allows imaging on a single focal plane. The magnet is removed from the sample and illuminated with fluorescent light emitted by the target cells captured by a CCD camera. In another embodiment, a floater having a permanent magnet makes the target cells array along a single imaging plane within the sample solution. An image analysis can be performed with a novel algorithm for measuring cells on the surface reflecting the target cell concentration of the original sample.

The method comprises (a) preparing cell fractions from a biological sample obtained from a test object in an immunology magnetic manner, where the sample includes a mixed cell population which has the possibility of containing cancer cells and includes a large amount of disturbing substances, and enriching the cell fractions for cancer cells if the cancer cells exist in the biological sample, and (b) analyzing such enriched fractions for the presence of the cancer cells, where the presence of cancer cells in the sample indicates the existence of incipient cancer in the test object.
