摘要： A sample containing an angiotensin-converting enzyme (ACE) is mixed with hippuryl-glycyl-glycine or hippuryl-L-histidyl-L-leucine as substrates, and is deproteinized and the released amount of glycil-glycine, respectively L-histidyl-L-leucine, is determined. Such determination is preferably carried out by arylation by means of nitrobenzin derivatives such as for example trinitrobenzinsulfonic acid or 1-fluoro-2, 4-dinitrobenzin and the arylated products are determined by photometry. A reagent for implementing such process and containing as ACE substrates hippuryl-glycil-glycine or hippuryl-L-histidyl-L-leucine is also disclosed.

摘要： Fluorogenic substrates for proteolytic enzymes having the formula: (FORMULA) or acid salts thereof wherein: R1 is hydrogen-L, hydrogen-D, benzoyl, benzenesulfonyl, glutaryl, pyroglutamyl, carbobenzoxy, D-serine, or carbobenzoxyserine; R2 is hydrogen, phenyl, a straight, branched or cyclic alkyl having 1 to 4 carbons, or propionic acid; R3 is hydrogen, straight or branched or cyclic alkyl having 1 to 4 carbons, 4-aminobutane, or 3-guanidylpropane; R4 is methyl, 4-aminobutane, or 3-quanidylpropane; R5 is a fluorogenic moiety, severable from said compound by a proteolytic enzyme and having different fluorescent property when severed from said compound than when forming part of said compound. The enzymes, when reacting with the substrate, remove the fluorogenic group R5 producing an increase in its fluorescence. The increase of the fluorescence is an indication of the activity of the enzyme present. Specific enzymes thusly detectable include plasmin, thrombin, the factors Xa, XIa, & XIIa, kallikrein, trypsin, elastase, urokinase, and cathepsin B1.

摘要： Improvements in methods for determining the presence of suprathreshold amounts of aspartate aminotransferase in an oral fluid sample by (1) incubation of aspartate aminotransferase substrate materials in a first reaction mixture with the sample to form a first reaction product and (2) detection of reaction product therein by incubation with an indicator substance in a second reaction mixture wherein a second colored reaction product is formed. The improvement comprises (a) utilizing a diazonium dyestuff as the indicator substance in the second reaction and (b) modifying reaction conditions within the first and second reaction mixtures such that no visually detectable colored reaction product will be formed in the second reaction mixture by subthreshold level of aspartate aminotransferase but wherein a visually detectable color reaction product will be formed in the reaction mixture when a selected suprathreshold concentration of aspartate aminotransferase is present in the first reaction mixture.
