摘要： A microfluidic flow synthesis method for collagen-based microparticles. Macro-porous fibrillar microparticles of pure collagen and composite collagen-GAG were formed, where the particle size and porosity were controlled by the flow synthesis parameters such as precursor flow rates. Using human dermal fibroblasts (hDFB) and umbilical cord derives mesenchymal stem cells (ucMSC), the capacity of the presented collagen and collagen-GAG microparticles to serve as cell culture substrates has been shown, to enable casting of tissues, forming spheroids, and as shear-thinning bioinks for bioprinting. At high packing densities, jammed collagen-based microparticles along with cells constitute a shear-thinning bioink for extrusion bioprinting applications. Bioprinted constructs exhibit a high porosity matrix due to the space between the microparticles, facilitating macromolecular transport, and cell proliferation and migration.

摘要： Centrifugal microfluidic fractionation is facilitated by off-chip pressurized fluid control applied at ports of a centrifugal microfluidic chip during centrifugation. The off-chip control is leveraged to align a desired fraction of the fractionated sample in a column on the chip to an extraction channel for greater precision extraction. Higher density or lower density media can be used to displace the fractionated sample axis-proximally or axis-distally, respectively. Automation by pixel-based strobed imaging of the chip with image analysis can reduce human intervention in fractionation assaying.

摘要： A centrifugal microfluidic device is provided having a microfluidic circuit, a fluid reservoir for providing fluid in the microfluidic circuit, a hydrodynamic resistance element in fluid communication with the reservoir for controlling rate of flow of a fluid out of the reservoir, and a siphoned chamber in fluid communication with the hydrodynamic resistance element and the microfluidic circuit for receiving fluid from the hydrodynamic resistance element and for delaying and metering of the fluid into the microfluidic circuit. The microfluidic device is useful for performing a biological assay. Operation of the device is completely independent on the liquid-solid contact angle and wetting properties of the liquids on the solid material of the platform, and the device does not need a carefully controlled rotation protocol.

摘要： The invention provides a new process for patterning TPE membranes for use in the design and fabrication of 3D microfluidic devices. The process involves patterning a TPE material without permitting the highest features of the mold to come into contact with the counter-plate, whereby adhesion between the TPE and the mold or counter-plate during demolding results directly in removal of the excess layer from the TPE membrane to produce well formed micrometric-sized open-through holes in the TPE membrane. The process permits rapid, reliable and efficient patterning of densely packed and arbitrarily placed micrometric open-through holes and channels of high aspect-ratio and any shape or wall profile in thin TPE membranes.

摘要： A technique for detection of probes in a microfluidic flow-through chamber involves a plurality of interface pinning reaction vessel formed by micro- or nano-structured relief patterning of a substrate. The relief patterning increases a surface area locally, and defines a plurality of separated interface pinning reaction vessels. The marked detection protocol may be supplied on a single layer of a stacked microfluidic chip, or the chamber may constitute a whole layer. The chip may be designed to be driven mechanically, pneumatically, hydraulically, centrifugally or by capillary action. Each vessel allows for a high density of probes, an effective region for developer-type or fluorescence-based marking, and efficient readout. Suitable probe liquids can be self-limiting to fill one vessel. Suitable developer liquids avoid dye bleeding across vessels during washing.

摘要： A technique for producing an artificial biointerface involves providing a patterned microfluidic chip having: a chamber divided by a fluid-permeable fencing into a central region and two flanking channels; and at least 3 fluid paths, each of the paths extending across one of the central region and the two flanking channels. A porous packing of rigid beads is placed within the central region to define a bead bed, the beads being of a size to be retained by the fencing. A biowall can be grown on at least one segment of the fencing separating the central region from one flanking channel, the biowall formed at least in part by live cells cultured on the beads. Beads may be modified, coated or functionalized to improve cell attachment and growth, and for reporting, or dosing particles or molecules can be conveniently added to the bead bed.
