It has been discovered that a novel dye, presently known as basic blue 93, a basic cationic azo dyestuff whose chemical structure is not of public record, but which is publicly available and accurately identified by its absorbance spectrum made a part hereof, when reacted with dried or fixed preparations of specimens of human blood and bone marrow cells reacts with lysosomes (primary granules) in the cells of the neutrophilic granulocytic series to provide novel diagnostic advances permitting differentiation between myeloblasts and lymphoblasts, for example, prior to specific therapy election in leukemic patients.

摘要： Differentiation, identification and enumeration of sub-populations of lymphocytes including B-cells and T-cells can now be further discriminated into their sub-populations by means of basic orange 21 as a supravital stain to provide distinctive morphologic differences between B-cells, T-suppressor cells, T-helper cells and natural killer (NK) cells without resorting to the use of individual monoclonal antibodies for differentiation of each of the above. The differentiation, identification and enumeration of the foregoing cells can be accomplished by means of both white light absorbance and fluorescence measurements, singly or in sequential combinations.

摘要： The present invention provides an advance in the art of cytology of blood, bone marrow and like tissue cell studies disclosing unique qualities and abilities of an unusual single cationic sulphur containing azo textile dye which in dilute aqueous solution stains identifying components present in the fixed human biopsy specimens after said specimens have been stained in a variety of different but reproducibly identifying metachromatic colors and patterns of colors having unusual color stain stability over extended time periods. Basic blue 41 makes possible, not only differentiation and identification of all the previously known blood cells identified by the Romanowsky-type dyes collectively in present day use but has been more recently established to differentiate and identify megakaryocytes, sub-populations of lymphocytes, including: B-cells, T-helper cells, T-suppressor cells, Natural Killer or NK cells, as well as blood parasites characteristic of certain tropical diseases, illustratively, Plasmodium Falciparum and Trypanosomiasis.
