摘要： Advancements in anti-viral therapies have led to long-term suppression of virus in infected patients. The suppressed virus can be maintained at levels that are undetectable in most assays. Although the virus cannot be detected, replication continues in the patient and the virus is able to acquire mutations that confer resistance to drugs in the anti-viral therapy. To improve treatment of patients whose virus accumulates resistance to drugs more sensitive assays are needed that can detect and genotype suppressed virus. Detecting viral nucleic acids in whole blood or that is associated with red blood cells increases the sensitivity of viral nucleic acid detection methods and increases the ability to obtain sequence information from the viral nucleic acids. This information is useful for evaluating anti-viral treatment regimens and providing more effective treatments to virally infected patients.

摘要： Testing methods that detect virus in donor samples have increased the safety of the blood supply and improved the ability of physicians to monitor virus replication in infected patients. These methods, however, are unable to detect virus in donor samples if the donor is tested immediately following infection or if the donor has low levels of virus present in the sample. Testing for the presence of virus in whole blood or red blood cells will increase the sensitivity of viral testing methods and expand the periods of time following infection at which virus can be detected.

摘要： The present invention is directed to methods, assays and compositions for implementing such methods and assays for assessing efficacy of individual components in multi-component vaccines and for assessing efficacy of a vaccine against a pathogen. In one aspect, the method of assessing efficacy of a vaccine against a pathogen is a quick assay that tests for an activity correlated with efficacy such as binding in an ELISA rather than requiring the time and expense of an assay that detects actual bactericidal activity. In another aspect, the method for testing the efficacy of an individual component in a multi-component vaccine includes obtaining an immune sample from a subject inoculated with the multi-component vaccine; blocking the portion of the immune sample that recognizes the individual component such as by addition of the individual component, and testing the efficacy of the immune sample to respond to the pathogen.
