摘要： The present invention provides method of detecting a predetermined biological activity. The method includes using an aqueous mixture comprising a first indicator reagent with a first absorption spectrum and a second indicator reagent. The second indicator reagent is converted by the predetermined biological activity to a second biological derivative with a second emission spectrum. The first absorbance spectrum includes detectable absorbance in at least a portion of wavelengths present in the second emission spectrum. The first indicator reagent is received and concentrated from an aqueous liquid by a substrate, facilitating the detection of the second biological derivative.

摘要： A method and system are provided for the selective use of deconvolution to reduce crosstalk between features of an image. The method to select areas of an image for deconvolution comprising the steps of: a) providing an image comprising a plurality of features, wherein each feature is associated with at least one value (v); b) identifying a test feature which is a high-value feature adjacent to a known low-value zone of the image, wherein the test feature has a tail ratio (r t ), which is the ratio of the value of the test feature (v t ) to the value of the adjacent low-value zone of the image (v o ); c) calculating a threshold value t which is a function of tail ratio (r t ) of the test feature; and d) identifying selected areas of the image, the selected areas being those where the ratio of values (v) between adjacent features is greater than said threshold value (T(r t )). Typically, the method of the present invention additionally comprises the step of deconvolving the selected areas of the image.

摘要： Systems and methods for detecting an analyte of interest in a sample. The system can include a first container comprising a filter portion. The filter portion can include a filter comprising a filtrand of the sample on a first side of the filter. The system can further include a second container comprising the filter portion coupled to a detection portion comprising a microstructured surface. The method can include providing the first container, coupling the filter portion to the detection portion to form the second container where the first side of the filter faces the microstructured surface, and centrifuging the second container toward the microstructured surface. The method can further include inverting the second container after centrifuging to decant the supernatant from the detection portion, such that a concentrate comprising a sediment of the sample is retained in the microstructured surface, which can be interrogated for an analyte of interest.

摘要： Compositions that comprise water, a first indicator reagent that can be converted by a first biological activity to a first biological derivative, and a plurality of particles are provided. The first indicator reagent can comprise a fluorogenic enzyme substrate having a fluorophore selected from the group consisting of umbelliferone, 7-aminocoumarin, β-naphthylamine, β- naphthol, fluorescein, resorufin, 9H-(l,3-dichloro-9,9-dimethyl acridin-2-one), rhodamine 110, a derivative of any of the foregoing fluorophores, and a combination of any of the foregoing fluorophores. The particles are capable of receiving and retaining the first biological derivative from an aqueous liquid. The first biological derivative can be indicative of a microorganism. The compositions further can comprise a gelling agent. Methods of using the compositions to detect the presence or absence of a microorganism in a sample are also provided.

摘要： Devices, methods and systems for low volume microarray processing are disclosed. The microarray devices preferably include a plurality of reactant sites on a reactant surface. The reactant sites include reactants that operate to capture one or more selected analytes that can then be detected based on an electromagnetic signal, e.g., fluorescence, that is emitted by each analyte in response to excitation energy incident on the microarray device. Mixing and/or distribution of the analyte sample over the reactant surface is accomplished by tilting the reactant surface such that the analyte sample flows over the reactant surface under the force of gravity. The tilting is performed such that a portion of the analyte sample accumulates in a bead along a first edge of the reactant surface. The reactant surface is then tilted in a different direction such that a portion of the analyte sample flows over the reactant surface and accumulates at a second edge. The reactant surface preferably generates sufficient capillary forces with the analyte sample such that the analyte sample is retained on the reactant surface. The tilting and resultant flow may be performed as many times as necessary to obtain the desired mixing and/or distribution of analyte sample over the reactant surface.
