    • The present invention relates to a novel peptide-N-glycosidase (PNGase Y) protein derived from Yarrowia lipolytica, to nucleic acids encoding said protein, to a recombinant vector comprising said nucleic acids, to a transformant transformed by said recombinant vector, to a method for preparing said protein by culturing said transformant, and to a method for preparing a glycoprotein in which an N-linked sugar chain is cut out using said protein or culture medium of said transformant comprising said protein. The peptide-N-glycosidase of the present invention may effectively cut out complex-type sugar chains, hybrid-type sugar chains, high-mannose-type sugar chains, sugar chains having mannose-6-phosphoric acids added thereto, sugar chains in which á(1,3)-fucose is added to a first N-acetylglucosamine, etc., and therefore exhibits wide and superior substrate specificity to sugar chains derived from animals, plants, or insects. The peptide-N-glycosidase of the present invention may cut out N-linked sugar chains not only when a glycoprotein is in a peptide state, but also when a protein is simply in a denatured state, and therefore may cut out N-linked sugar chains in a more convenient and quicker manner and enable the analysis of the structures of the sugar chains.
