摘要： Various methods are described that provide for high resolution melt (HRM) genotyping, Some embodiments comprise providing a locus specific primer, and two allele specific primers each comprising at least one single nucleotide polymorphism (SNP) allele-hybridizable sequence, wherein at least one of the allele specific primers also comprises at least one nucleotide alteration. In some embodiments, a nucleic acid is provided comprising a SNP base located within 1-20 bases of its 3' end. Some embodiments comprise hybridizing the locus specific primer and at least one of the allele specific primers to the nucleic acid, amplifying the hybridized nucleic acid using pyrophosphorolysis activated polymerization (PAP) PCR, and determining the melting temperature (Tm) of the resulting amplicons, for example, using HRM. In some embodiments, reaction mixtures and kits for HUM genotyping are provided. The reaction mixtures and kits can each comprise a locus specific primer, one or more allele specific primers each comprising at least one SNP allele-hybridizable sequence, and a PAP PCR enzyme, wherein at least one of the allele specific primers also comprises a nucleotide alteration, for example, a tail.

摘要： An assay card and devices and methods for isolating chambers on the assay card are described. The assay card comprises a substrate formed of one or more materials, e.g., plastic, having a softening temperature, the substrate defining channels communicating with respective reaction chambers. The assay card may be heated in a region of the channels to at least the softening temperature. The softened plastic may be deformed, e.g., with a tool which may or may not also provide the heat for softening the substrate. In this manner, the plastic of the substrate may be caused to at least partially obstruct the channels, thereby isolating the reaction chambers. The invention also relates to a method of manufacturing a tool device that includes pins for heating and deforming an assay card.

摘要： Modification formats having modified nucleotides are provided for siRNA. Short interfering RNA having modification formats and modified nucleotides provided herein reduce off-target effects in RNA interference of endogenous genes. Further modification formatted siRNAs are demonstrated to be stabilized to nuclease-rich environments. Unexpectedly, increasing or maintaining strand bias, while necessary to maintain potency for endogenous RNA interference, is not sufficient for reducing off-target effects in cell biology assays.
