摘要： A method of forming an amino acid thiohydantoin from an N-protected amino acid or the C-terminal amino acid of an N-protected peptide. The amino acid is activated by reaction with a ketenimine, and the activated ester is converted to the thiohydantoin by reaction with silyl or pyridine isothiocyanate. The ketenimine is generated by treating an N-substituted isoxazolium compound, such as Woodwards Reagent K with a base, preferably in the presence of the amino acid. Also disclosed is a solid phase support having a derivatized N-substituted isoxazolium or ketenimine group for use in the method.

摘要： A liquid-handling instrument (11) has a worksurface (22) with registration for modular stations to support containers of liquid, pipette apparatus with a pipette tip (33) coupled to a sensing circuit, a robotic translation system (31) for moving the pipette tip, and a control system with an iconic user interface for programming and editing. A gauge block (24) registered on the worksurface provides for calibration using the sensing tip, and register cavities on the worksurface provide for modular stations. There is a wash station (25) for the pipette tip on the worksurface. An automated laboratory based on the liquid-handling system has heating and cooling and a sealable incubation station (21) as well as a magnetic separation station (26 and 29). Methods are disclosed using the apparatus to convey droplets of liquid, to aspirate with minimum tip contamination, to mix liquids in containers, and to validate the worksurface. A duck-billed closure (253) is disclosed for minimizing evaporation and cross-contamination during processing, and is a part of a container disclosed for storing and transporting liquids.

A key feature in the preparation of the succinimidylcarboxylates is the use of stoichiometric amounts of di-N-succinimidylcarbonate and 4-dimethylaminopyridine for converting the 5- and/or 6-carboxyrhodamines to N-hydroxysuccinimide esters. In chain termination DNA sequencing procedures, using the pure isomeric forms of the activated rhodamines prevents generation of spurious sequence data because of the different electrophoretic mobilities of the isomers.

摘要： A device for mixing liquids contains a mixing chamber (1) into which a mixing head propelled around a rotationally axis is designed. Between the wall of the mixing chamber and the mixing head (2) a cleft mixing area is created. Through rotation of the mixing head (2) a shear flux of the injected liquids (11, 12, 13, 14) is created which causes mixing. With this active mixing method minute liquid amounts with differing volumes and viscosities can be mixed with high rapidity. The device is especially suitable for the automated analysis of enzyme kinetics.
