摘要： A modular apparatus for continuous production of purified RNA, the apparatus including, in operable communication: a reaction module including a bioreactor configured for continuous synthesis of an affinity-tagged RNA molecule by in vitro transcription using reactants in an in vitro transcription mixture; a control module including a sensor; and an RNA separation system configured to receive a transcription reaction product comprising the affinity-tagged RNA molecule from the bioreactor and to separate the affinity tagged RNA molecule from the transcription reaction product, wherein the RNA separation system includes a capture module including at least one affinity column for binding the affinity-tagged RNA molecule and a purification module including a continuous chromatography column, wherein the control module controls input of the reactants to the bioreactor and monitors separation of the affinity-tagged RNA molecule using the sensor, and wherein the sensor is in communication with the reaction module and the RNA separation system and the sensor provides feedback control of the transcription mixture in the reaction module and the transcription reaction product in the RNA separation system.

摘要： Methods for purifying RNA from a sample, comprising one or more steps of tangential flow filtration, hydroxyapatite chromatography, core bead flow-through chromatography, or any combinations thereof. These techniques are useful individually, but show very high efficiency when used in combination, or when performed in particular orders. The methods can purify RNA in a highly efficient manner without unduly compromising potency or stability, to provide compositions in which RNA is substantially cleared of contaminants. Moreover, they can be performed without the need for organic solvents.

摘要： A secondary plasma device (SPD) and methods for its use with apheresis machines or similar extracorporeal blood processing systems and capable of collecting cell-free nucleic acids (cfNA) directly from a subject's circulating plasma, the SPD comprising:

i. a sterile capsule having an inlet port and an outlet port and enclosing a filtration/adsorption medium, and configured to receive plasma from the extracorporeal blood processing circuit through the inlet port, such that plasma enters under pressure and flows through the filtration/adsorption medium then exits through the outlet port to a return path of the extracorporeal blood processing circuit leading back to the subject; and
ii. the filtration/adsorption medium bearing fixed positive charge that captures cfNA from plasma by anion exchange, and from which cfNA can be subsequently recovered in quantities 10 to 1000-fold greater than the amount of cfNA contained in a 5 ml venipuncture specimen of the subject's blood.

摘要： Provided is a linker for both screening assessment of the candidate clones without using enzymes, and to provide an in vitro selection method using thereof. Also, provided is a high-speed photo-crosslinking linker for molecular interaction analysis and in vitro selection comprising a backbone and a side chain. The backbone comprises a solid-phase binding site located at the 5′ terminus for forming a bond with a solid-phase; a solid-phase cleavage site for releasing the entire solid-phase at the site; a side chain linking site for linking a side chain; a high-speed photo-crosslinking site for linking the backbone to mRNA having a sequence complementary thereof via photo-crosslinking; and a reverse transcription initiation region located adjacent to the side chain linking site at the 3′ terminus of the backbone. The side chain comprises a fluorescent label, a protein binding site located at the free terminus thereof; and a binding site with the backbone.

Contacting the sample with a hydrophobic interaction chromatography (HIC) material in a solution comprising a kosmotropic salt in a concentration which forces the pDNA and contaminant to adsorb on the HIC material,
Diluting the concentration of the kosmotropic salt in presence of a neutral salt subsequent to adsorbing of the pDNA on the HIC material, thereby
Desorbing the pDNA from the HIC material, whereas the contaminant stays adsorbed by the continued presence of the neutral salt, and
Obtaining the pDNA preparation.

Furthermore, a method is disclosed for preparing a sample to be subjected to the method of the invention or other purification methods, in particular anion exchange chromatography by exposing the sample to a neutral salt in the presence of a HIC material.
