    • The present invention provides a preparation of botulinum toxin light chain type A or E, wherein the preparation is both catalytically active and soluble. Preferably, the preparation consists essentially of amino acid residues 1 through 425 of the botulinum toxin light chain type A. A method of screening inhibitors is also provided, wherein the method comprises exposing a test inhibitor to the preparation of botulinum toxin light chain type A and evaluating the biological activity of the preparation. In another embodiment, a method of providing a catalytically active, soluble preparation of botulinum toxin light chain, type A is provided, wherein the method comprises obtaining an expression vector comprising a DNA sequence encoding amino acid residues 1-425 and expressing a polypeptide.

    • A method in a mobile device for use in receiving updates of network-maintained map data sets is described. The mobile device receives, through a wireless network, map data for rendering a map. The map data has a plurality of layers, where each of the plurality of layers includes a version number and a plurality of data entries associated therewith. Each of the plurality of data entries indicates a plurality of data points for rendering one or more artifacts or labels within an area of interest of the map. The mobile device stores the map data, and displays the area of interest from the plurality of layers of the stored map data. The mobile device then receives, through the wireless network, an indication of an alternative version of one of the layers of the stored map data. When the version number of the alternative version is greater than the version number of the stored version, the mobile device causes one or more requests for the alternative version of said layer for the area of interest to be sent through the wireless network. The mobile device also receives the alternative version of said layer for the area of interest through the wireless network in response to the one or more requests. The mobile device also then causes the map of the area of interest to be rendered in the display in accordance with the combined plurality of layers of the stored map data, but with use of the alternative version of said layer.

    • This is directed to controlling the output of audio based on the mode of an electronic device. In particular, this is directed to an electronic device having a sound mode, silent mode, and mute mode controlling the output of audio along different audio output routes. In the sound mode, all audio can be output, while only registered or authorized audio can be output in the silent mode. In the mute mode, no audio can be output. The sound and silent modes can be enabled using an interface of the device (e.g., a switch having two possible states), while the mute mode can be enabled using an accessory coupled to the device. To disengage the mute mode, a user can provide a corresponding instruction using the accessory, or providing an instruction on the device related to volume control. For example, a user can change the device volume using an on-device interface, or toggle a sound/silent switch to a sound mode.

    • A downhole valve has a closure device (e.g., one or more flappers) for closing off the valve. A no-go actuation mechanism protects the flappers from damage. When the flappers are closed, the mechanism prevents a tool from passing into the valve and causing damage to the flappers. Yet, the mechanism may open the valve's flappers when the tool string is forced into the valve. When the valve has successfully opened, then the mechanism moves out of the way of the toolstring so it can pass through the valve. For the mechanically operated valves, operators use a shifting profile in the valve only in the upward direction to return the valve to the closed position. For hydraulic actuated valves, hydraulic pressure may be used or exhausted, depending on the design, to allow the flappers to go closed. Once the flappers have closed, the no-go mechanism is once again realized.
