    • Signals obtained in High Performance Liquid Chromatography (HPLC) are notoriously noisy. The signal can be improved by increasing the path length of the light passing through the sample, but increased path length also invites greater refraction and reflection of that light. A novel approach is to utilize a number of photocell sensors lined up along a capillary through which the solution passes. Through careful measurement and calculation, a signal for a single fluid particle can be produced by all the photocell sensors. By incorporating an appropriate delay between each signal, to take into account the finite speed of the fluid, the resulting plurality of noisy signals can be summed or statistically correlated. The signal, containing the important information about the fluid, will increase with the number of photocells used, while the noise increases by the square root of the number of photocells used. The signal to noise ratio, then, increases as the square of the number of photocells used. In this way, the effective path length is increased without including any moving parts.

    • A high dynamic range apparatus for separation and detection of polynucleotide fragments has a housing adapted to receive an electrophoresis gel holder containing an electrophoresis gel loaded with fluorophore-labeled samples; one or more laser diodes for providing radiation of a frequency suitable for excitation of the fluorophore which irradiates a an array of excitation/detection sites on the electrophoresis gel; an array of detectors aligned with the excitation/detection sites for collecting fluorescent emissions; and one or more components for increasing the dynamic range of the instrument by at least an order of magnitude. These components, which can be used individually or in combination include detectors that are connected to a signal processing system that modulates the period of signal integration employed so that large signals are totaled at short time intervals and smaller signals are totaled at longer time intervals; the use of a beam splitter to produces a high intensity beam of emitted light and a low intensity beam of emitted light from each excitation/detection site; and a device for modulating the intensity of the excitation beam can be used to improve the dynamic range of the instrument.

    • A method of analyzing data from a mass spectrometer for a data dependent acquisition is described. In an embodiment of this method, mass spectral scans are taken of a sample eluted from a liquid chromatography column. An extracted ion chromatogram (XIC) is then created for each m/z data point of the mass spectral scans and the XIC for each m/z data point are correlated to a model function, such as a monotonically increasing function, or the first half of a gaussian function, to obtain a XIC correlation value. A weighting function is then applied to the XIC correlation value to obtain a current weighted intensity. The current weighted intensity for each m/z point is used to reconstruct a weighted mass spectrum, which is then used to make a real-time decision for the data dependent acquisition. In an embodiment, the data dependent acquisition is the performance of tandem mass spectrometry. Embodiments of the invention also describe a sample processing apparatus for the data dependent acquisition and a computer readable medium that provides instructions to the sample processing apparatus to perform the method described above.
