    • PURPOSE: Provided is a method for manufacturing 5-demethylovalicin which selectively inhibits the activation of MetAP2 and shows great effect on preventing angiogenesis so that the compound is very useful in manufacturing medications for cancer, rheumatoid arthritis, and diabetes. CONSTITUTION: A method for manufacturing 5-demethylovalicin of the formula (1) comprises the following steps: i) selecting and identifying Chrysosporium lucknowense; ii) inoculating a colony on PDA media (potato dextrose agar) into YM medium and cultivating the colony at 26 deg.C at 130rpm for three days; iii) inoculating 2-3%v/v of the broth from step (ii) into medium containing 0.5-3.0% of glucose, 0.2-0.5% of yeast extract, 0.5-1.5% of peptone, 0.1-0.5% of KH2PO4, and 0.05% of MgSO47H and cultivating the broth at 25-28 deg.C at 250rpm for six days; iv) fermenting the broth at 180-240 rpm for 4-6 days; v) obtaining extracts by mixing 16 L of the fermentation broth with the same amount of acetone; vi) decompressing and condensing the mixture to eliminate acetone and get second extracts; vii) loading the second extracts on silica gel and decompressing and condensing activated segments; and viii) performing Prep-TLC with the activated segments to obtain purified compounds.
