    • A peptide nucleic acid(PNA) oligomer including at least one universal base is provided to be applied to a PNA microarray and a PNA chip as a probe, thereby detecting a nucleic acid including SNP(single nucleotide polymorphism) and mutation with high specificity. A PNA oligomer consists of a base sequence represented by a formula(1) of Ap-Bq-Cr and is characterized in that it has increased solubility in water and specificity upon hybridization with a nucleic acid by introducing at least one universal base capable of forming a base pair with natural DNA or RNA bases into the base sequences having at least 60% purine bases or at least four contiguous purine bases. In the formula(1), each Ap and Cr is a base sequence complementary to a base sequence other than a specific base sequence of a target nucleic acid; B is a base sequence complementary to a specific base sequence of a target nucleic acid, wherein the target nucleic acid includes the specific base sequence containing at least 60% of pyrimidine bases or at least four contiguous pyrimidine bases; each p, q, and r is a number of the bases, p is an integer from o-15, r is an integer from 0-15, and q is an integer from 1-30, provided that p+q+r is an integer from 10-30; and Ap-Bq-Cr is a base sequence to which at least one universal base, capable of forming a base pair with natural DNA or RNA bases, is introduced, sequences selected from SEQ ID Nos are 2 to 7. A method of preparing the PNA oligomer comprises the steps of: investigating whether a PNA oligomer comprises at least 60 bases or at least four contiguous purine bases in its base sequence or not; (b) synthesizing PNA monomers comprising natural bases and universal bases, respectively; and (c) introducing at least one universal base, capable of forming a base pair with natural DNA or RNA bases, to the base sequence with the PNA monomer, to prepare the PNA oligomer. A kit for analyzing, detecting or modulating nucleic acid comprises the PNA oligomer. A device for analyzing or detecting nucleic acid comprises the PNA oligomer.

A method for preparing the compound of formula (1) comprises reacting a compound of formula (5) with a compound of formula (6), wherein the compound of formula (5) is prepared by (a) reacting a compound of formula (6) with a compound of formula (2) to prepare a compound of formula (3), (b) hydrolysis of the compound of formula (3) and reacting it with di-t-butyl dicarbonate to prepare a compound of formula (4), and (c) cyclization of the compound of formula (4) and deprotecting the compound.
