The application of voltage to the pump becomes reverse to separation electrophoresis, that is, becomes a cathode 11 and an anode 12 but a separation cathode 9 and the pump cathode 11 are used in common to make it possible to form even a three- pole structure. The preparatory data from the auxiliary detector 20 provided to a separation capillary migration part other than detector 4 is fed back to an electrophretic condition controller 21 to be used in the control of separation or pump applied voltage.

    • PURPOSE:To detect the minute difference between protein mixtures sensitively by obtaining the product of the three values, i.e. the sum of absorbances, the changing rate of the area and the deviation of the central position of the chromatin spot of the protein component wherein two two-dimensional electrophoresis images obtained under the same migrating conditions are segregated. CONSTITUTION:Rhodamine is injected into a rat, and a rhodamine sarcoma is developed. The sarcoma is transplanted into the back part of another DONRYU rat. After the transplant, the serum protein is analyzed by an electrphoresis method with the elapse of time. An electrophoresis image wherein the spot of the segregated protein clearly appears is formed. The image data are inputted into a computer through a TV camera, and the image is analyzed. Namely, the sum of the absorbances, the change in area and the deviation of the central position of the chromatin spot are obtained for every segregated spot, and the product of the value is computed. Thus, the change in serum protein due to the sarcoma is detected with good sensitivity.

    • PURPOSE:To achieve automation of a substrate treating stage and a dyeing stage by preparing a treating tank, treating liquid tank and waste liquid tanks and connecting these tanks via respective solenoid valves by means of pipings and changing over the solenoid valves to conduct the treating liquid to the treating tank. CONSTITUTION:Substrates 2 are disposed in a treating tank 1 and four solenoid valves 3 and the same number of treating liquid tanks 4 are provided above the same and are connected respectively by a piping 5. Four solenoid valves 6 and waste liquid tanks 7 are provided below the tank 1 and are connected to each other similarly by a piping 10. These valves 3, 6 are controlled by a controller 8. The treating liquid 9 contained in the tanks 4 is fed to the tank 1 by opening and closing of the valves 3 and the substrates 2 are treated. The valves 6 are opened upon ending of the treatment to conduct the liquid 9 to the prescribed tanks 7. The similar operation is repeatedly performed with the other treating liquid as well.

    • PURPOSE:To improve a separating power sharply by using a plane-shaped gel having the concentration gradient of polyacrylamide and containing a buffer liquid having alkaline hydrogen ion concentration from an isoelectric point of a component to be inspected. CONSTITUTION:One surface of a glass plate 1 is processed by methacryl oxypropyltrimethoxysilane, and thereby a plane gel 2 having the concentration gradient of 4-20% and containing a buffer liquid (PH-8.6, ion strength-0.06, A liquid) composed of 5,5-diethylbarbituric acid sodium and 5,5-diethylbarbituric acid is prepared. A serum is given into a cut 3 made in the low-concentration part of the gel. Filter paper sheets 4 and 5 for liquid junction, which are impregnated with the A liquid, are connected to a negative electrode tank 6 and a positive electrode tank 7 with the A liquid put therein, and serum protein is separated by electrophoresis. The liuid junction being removed, filter paper sheets 8 and 9 for liquid junction, which are impregnated with the A liquid, are connected to a negative electrode tank 10 and a positive electrode tank 11 with the A liquid put therein, and the serum protein is separated according to the difference in molecular weight.

    • PURPOSE:To carry out two-dimentional cataphoresis using the same plate gel and to improve analytical method of blood serum protein etc., by giving the concentration gradient of polymer gel in one direction of the plate and using the plate gel added amphoteric electrolyte to the lower concentration part. CONSTITUTION:Polymer gel, such as acrylamide etc., having molecular sieve effect, is formed on the plate put in the vessel at about 2.5-40% of concentration gradient. About 0.5-2wt% of amphoteric electrolyte composed of reaction product etc. of amine, such as triethyleneteramine etc., having secondary amino radical and alpha, beta unsaturated acid, such as acrylic acid etc., is added to diluted side of the concentration gradient. Electrodes are provided at both ends of the plate and the sample to be tested is covered on the diluted part of polymer gel concentration and then, the one-dimentional cataphoresis is carried out in a constant direction of the polymer gel. Next, two-dimentional cataphoresis is carried out in the concentration gradient direction of the polymer gel. By the above method, automation of the apparatus is able to carry out.
