摘要： PURPOSE: To obtain a new deacetylase important for producing a deacetylated substance of an N-acetylchitobiose and an N-acetylchitotriose useful as a synthetic intermediate for medicines, etc. CONSTITUTION: This deacetylase DA1 or DA2 is obtained by culturing a Vibrio sp. H-8 strain (FERM P-14737), etc., and having the following properties: (l) each capable of acting on an N-acetylchitobiose or an N-acetylchitotriose and carrying out the deacetylation thereof; (2) optimal pH8.5-9.0 (37 deg.C), 45 deg.C optimal temperature (pH8.5), stable by treatment at pH8.5 and 50 deg.C for 1hr, 20000-60000 molecular weight [measured according to a sodium dodecyl sulfate- polyacrylamide gel electrophoretic method (SDS-PAGE)] and 3.3 isoelectric point for the deacetylase DA1; (3) optimal pH8.0-8.5 (37 deg.C), 40 deg.C optimal temperature (pH8.5), stable by treatment at pH8.5 and 50 deg.C for 1hr, 20,000-60,000 molecular weight (measured according to the SDS-PAGE method] and 3.5 isoelectric point for the deacetylase DA2 and (4) each having high activities.

摘要： PURPOSE: To obtain a new enzyme-inhibiting substance having a chitinase- inhibiting activity and useful for antifungal agents, agricultural chemicals, etc., by culturing a microorganism belonging to the genus Pseudomonas and having a cyclo-L-arginyl-D-proline-producing ability in a culture medium. CONSTITUTION: This new substance is cyclo-L-arginyl-D-proline expressed by formula. The substance has a chitinase-inhibiting activity, inhibits the chitinase for controlling the metabolism of chitin important for ecdysis and growth of crustaceans, insects, etc., or the division and multiplication of molds, enzymes, etc., and can be utilized for agricultural chemicals such as an antifungal agent, an insect growth-controlling agent, etc. The compound is obtained by inoculating a culture medium with a microorganism [e.g. Pseudomonas SP.IZ 208 strain (FERM P-14738)] belonging to the genus Pseudomonas and having a cyclo-L- arginyl-D-proline-producing ability as a spwan, subjecting the inoculated microorganism to a shaking culture at 25 deg.C for 72hrs, centrifuging the culture solution, collecting the supernatant, separating the product from the collected supernatant, and subsequently purifying the product.

Thereby, the chitinase-inhibitor is simply assayed with high measuring sensitivity and it is useful for development, etc., of a method using chitinase-inhibitor capable of controlling growth of agricultural insert pests.

摘要： PURPOSE: To obtain a novel chitinase inhibitor which is useful as an antimycotic agent, agrochemical or the like by using a cyclic dipeptide containing an arginine residue having high inhibitory activity against chitinase as an active ingredient. CONSTITUTION: A cyclic dipeptide which contains arginine residue and is represented as a diketopiperazine derivative by formula I or II (R1 is a side chain originating from the amino acid, for example, H in glycine, methyl in alanine or isopropyl in valine; R2 , R3 are each H, OH) is used as an active ingredient to give this inhibitor. The dipeptide content is preferably 0.1-70wt.% based on the whole preparation. This cyclic dipeptide of formula I or II is readily produced, for example, by effecting the condensation reaction between protected arginine and another amino acid using N,N-dicyclohexyl-carbodiimide, followed by deprotection and cyclization. The objective chitinase inhibitor is useful as a new type of antimycotic agent, a growth regulator of insects and a therapeutic agent for candidiasis.
