      CONSTITUTION: A microorganism having the aimed gene, vector and promoter in the cell and an expression ability of the aimed gene is cultivated in a culture medium containing an inhibitor substance and inducer substance to control the promoter activity and give the product of the aimed gene efficiently in a large amount. For example, a DNA fragment containing a trp promoter cut out of PTREI is linked to an β-gal gene with an enzyme to create a complex plasmid PTREZI, and tryptophan (trp) is added thereto in the initial period of cultivation to suppress the activity of the trp promoter, reduce the production of the β-gal and multiply the microbial cells. An inducer substance for starting the activity of the trp promoter is then added to produce the β-gal.

The controlling part 6 is furnished with the pipes 8, 10 to transfer the reaction liquid flowed down from the upstream-side reaction layer 5 to the downstream-side reaction layer 5. A mixing part is placed between the pipes 8 and 10, and an inlet 11 of a pH-controlling liquid is attached in the mixing part 9, and the inlet 13 and the outlet 14 of the medium for the temperature control of the reaction liquid is attached to the mixing part 9.

    • PURPOSE:To process waste water econimically with out adding organic substances as nourishment by effecting positive ion-exchange for a part of waste water including organic substances and nitrogen oxide and introducing processed water into denitrifying process while positive ion-exchange substances into nitrifying process. CONSTITUTION:When a part of waste water 1 including organic substances and ammonium, nitrous or nitric nitrogen is sent to a positive ion-exchange process tank 7 and is contacted with positive ion-exchange substances such as zeolit, the ammonium nitrogen in the waste water 1 is removed by positive ion-exchange. Thereafter, this processed water is sent to a denitrifying tank 5 through a settling tank 9 while the substance, which absorbed the ammonium nitrogen separated in the settling tank 9 and procesed by the positive ion exchange, is sent to a nitrifying tank 4. Then, denitrifying bacteria are activated by nourishment of organic substances in the processed water in the denitrifying tank 5 and the positive ion-exchange substances in the nitrifying tank 4 are revived and circulated to the positive ion-exchange process tank 7 and thus reused.
