SOLUTION: The method for counting and analyzing the residual leucocytes in a sample of a blood product selected from a group of plasma, erythrocytes, and platelets and having a reduced number of leucocytes includes a process (a) for forming a suspension of the blood product having a reduced number of leucocytes by bringing the blood product having a reduced number of leucocytes into contact with a sufficient quantity of a dissolving reagent, which contains both a low molar concentration of a buffer agent (i) and a nonionic surface-active agent (ii); a process (b) for incubating the suspension of the product having a reduced number of leucocytes and the dissolving reagent at temperatures sufficient for selectively dissolving platelets and erythrocytes without damaging the leucocytes over a sufficient period of time; a process (c) for bringing the leucocytes of the dissolved blood product into contact with pixels appropriate for dyeing the leucocytes; and a process (d) for measuring the number of the dyed leucocytes. 摘要： The invention provides a very fast and reliable method and reagent composition for determining reticulocyte and erythrocyte counts, identification and characterization, as well as platelet counts, in a whole blood sample using fully automated hematology analyzers. The reagent composition includes a zwitterionic surfactant as sphering agent for reticulocytes and erythrocytes, an organic cationic dye, e.g., Oxazine 750, for staining the reticulocytes, and buffer solution components for maintaining a reagent pH of about 7.2 to 7.8, and an osmolality of about 292 +/- 5. On the basis of the particular pH, ionic strength, and dye concentration, the reagent composition of the present method improves upon previous methods, thus allowing fully automated blood sample analyses to be completed in less than about 30 seconds, with only about a 20 second incubation of sample in the reagent solution before analysis. With such a rapid incubation time, the reagent mixture is passed through the sensing region of a flow cytometer, and the light scattered and absorbed by each cell is rapidly measured to provide accurate cell determinations and counts. The volume and hemoglobin concentration and content of each erythrocyte or reticulocyte are determined. Mean cell volume, mean corpuscular hemoglobin concentration, and mean cell hemoglobin of the reticulocytes and/or erythrocytes are calculated from the measured cell-by-cell volume and hemoglobin concentration.

摘要： The invention describes novel methods for calibrating or standardizing flow cytometry instruments using synthetic polymer particles or beads having physical properties which provide advantages for their use in such instruments. The polymer particles or beads employed in the calibration methods are spherical, have an average particle diameter of from about 1 to about 8 microns, a relatively narrow particle distribution and a low refractive index, i.e., from about 1.35 to about 1.45. The particles are safe, stable for long time periods and are particularly suitable for employment as hematology analyzer calibration substances. The invention provides an advantageous improvement over the types of standards currently used to calibrate a variety of instruments that rely on light scatter combined with other parameters to identify and measure biological particles or cells, as well as nonbiological particles. In particular, using the calibration/standardization method and polymer particles according to the present invention allows hematology instruments to be calibrated and standardized for subsequent accurate and reliable determinations of red blood cells, reticulocytes, white blood cells and platelets in whole blood samples.

摘要： The present invention provides a highly sensitive and accurate method and system for the discrimination and quantification of platelets in a whole blood sample using automated hematology instruments. The method and system of the invention provide the accurate measurements of platelet dry mass and platelet component concentration in both normal blood samples and in abnormal blood samples, such as those from thrombocytopenic patients. The determination of platelet dry mass and platelet component concentration can serve to assess the activation state of platelets since activated platelets possess measurably lower component concentrations and refractive indices than do unactivated platelets. The method and system of the invention also allows the clinician or skilled practitioner to determine the age of a blood sample on the basis of the measured parameter of platelet component concentration.
