    • An improved multi-purpose blood diluent for use with a gentle lysing agent and improved detergent reagent system are disclosed which are especially suitable for use in routine electronic enumeration and volumetric differentation of blood cells. The preferred imidazole stabilizer used in the diluent reagent is found to be an excellent cell stabilizing agent and buffer for maintaining cell morphology during operation. A synergistic combination of a superior antimicrobial agent, the preferred Triadine-10, used in the diluent and the detergent reagents, not only prevents bacterial or fungal growth, but also helps to stabilize cells and to obtain distinct volumetric differentiation of certain leukocyte subpopulations. The preferred Brij 35 in a balanced salt solution has proved to be an efficient and cost effective detergent to ensure accurate results and trouble-free operation of the analyzers.

    • Improved reagents and methods for obtaining distinct volumetric differentiation of platelets, erythocytes and certain leukocyute subpopulations are disclosed. 1,3-dimethylurea is disclosed as a cell stabilizing agent for use in the blood diluent reagent. A hematology analyzer detergent is described wherein the addition of a wetting agent to the diluent of the instant invention provides the necessary attributes of an automatic analyzer detergent. The lysing reagent exploits the selective and intrinsically gentle lytic qualities of the dodecyl quaternary ammonium salt. The improved method modifies the leukocytes so that the frequent lymphocyte and neutrophil subpopulations are well separated on a volume scale, permitting quantitative evaluation of infrequent and rare leukocyte subpopulations as well as improved enumeration of the lymphocytes and neutrophils.

    • A transfer valve system, method and apparatus are disclosed having a three element valve with a central element sandwiched between front and rear outer elements. Between an aspiration and a discharge position the front and rear outer elements are moved in opposite directions of unequal distances in the same direction with respect to the central member or one outer member and the central member are moved in the same or opposite direction with respect to the other outer member so long as the outer members end up shifted with respect to each other. In the aspiration position, two sample segments are contained in series within the valve. In one embodiment of the invention, one segment is contained within a conduit in the central member and the other segment is contained within a passageway in the rear member. In another embodiment of the invention, the two segments are contained within different conduits of the central member.
