The method comprises the steps of: (b) providing a lysed sample of whole blood; (b) introducing the lysed sample to a light-scattering multi-angle depolarizing flow cytometer; (c) removing depolarizing interference, e.g., lipid droplets and other measured particles; (d) differentiating nucleated red blood cells and noise from white blood cells in the absence of depolarizing interference; (e) differentiating nucleated red blood cells from noise in the absence of depolarizing interference and white blood cells; and (f) differentiating possible platelet clumps.

摘要： Cyanide-free reagents for the determination of hemoglobin and leukocytes present in a blood sample contain an aqueous solution of at least one quaternary ammonium salt, preferably selected from the group of alkyltrimethylammonium salts, alkyldimethylbenzylammonium salts or alkylpyridium salts consisting of tetradecyltrimethyl ammonium bromide (TTAB), dodecyltrimethyl ammonium chloride, cetyltrimethyl ammonium bromide, hexadecyltrimethyl ammonium bromide and benzalkonium chlorides, and hydroxylamine salts, especially hydrochloride, sulfate and phosphates and other acid salts. The method involves mixing the reagent with a diluent- diluted blood sample, presenting it to an absorbance spectrophotometer and measuring the resulting optical density as an indicator of hemoglobin concentration. This cyanide-free reagent could be used solely for hemoglobin determinations, or , it can also be used in leukocyte counting and sizing using hematology instrumentation.

摘要： An improved multi-purpose blood diluent for use with a gentle lysing agent and improved detergent reagent system are disclosed which are especially suitable for use in routine electronic enumeration and volumetric differentation of blood cells. The preferred imidazole stabilizer used in the diluent reagent is found to be an excellent cell stabilizing agent and buffer for maintaining cell morphology during operation. A synergistic combination of a superior antimicrobial agent, the preferred Triadine-10, used in the diluent and the detergent reagents, not only prevents bacterial or fungal growth, but also helps to stabilize cells and to obtain distinct volumetric differentiation of certain leukocyte subpopulations. The preferred Brij 35 in a balanced salt solution has proved to be an efficient and cost effective detergent to ensure accurate results and trouble-free operation of the analyzers.
