摘要： PROBLEM TO BE SOLVED: To provide a method for preserving a microbial membrane with which the activity of a microorganism can be maintained over a long period without dipping the microorganism-immobilizing membrane in a liquid.
SOLUTION: The method for preserving the microbial membrane comprises detecting a specific substance using respiratory inhibition of an iron bacterium as an index. Furthermore, the method comprises a step of immobilizing the microorganism 1 in the immobilizing membrane 2, a step of housing the immobilizing membrane 2 in a holder for fixing electrodes, a step of preparing a humectant carrier 3 soaked with a humectant liquid for preventing drying of the immobilizing membrane 2 and a step of housing the holder 7 for fixing the electrodes and the humectant carrier 3 in a hermetically sealed container preventing the entry of oxygen together with an inert gas in such a state that the microorganism 1 immobilized in the immobilizing membrane 2 housed in the holder 7 for fixing the electrodes is kept out of contact with the humectant carrier 3.

SOLUTION: The detection method is provided with: a step for capturing solid matter of a microorganism to be detected in sample water containing the microorganism; a step for fixing the captured solid matter onto a fixing plate; a step for bringing a labelling antibody, for specifically dying the microorganism to be detected, into contact with the solid matter on the fixing plate; a step for acquiring a two-dimensional image by irradiating light for sorting out the dyed solid matter; a step for forming double strands by in-situ hybridization using a nucleic acid probe, which has a base sequence complementary to nucleic acids contained in the microorganism to be detected and is labeled differently from the labeling antibody; a step for acquiring a two-dimensional image by irradiating light for sorting out the solid matter which forms the double strands; and a step for recognizing an image which appears in the same coordinates among the two-dimensional images as the microorganism to be detected.

Further, the apparatus is so structured that a labeled probe which is not hybridized is washed and removed after hybridization reaction is completed, a solution for releasing the probe is added thereto, the target nucleic acid which is hybridized and fastened to the beads is made to be settled on a bottom surface of a container, the labeled probe is released from the beads to a supernatant by gradually raising a temperature of a reaction solution up to a temperature higher than a denaturation temperature of the target nucleic acid, and a quantity of the fluorescent light or the luminescent light is successively measured.

SOLUTION: In a method of measuring the object substance in the sample by detecting the fluorescent or luminous labelled substance combined with the beads after a specific bond reaction using the beads is caused, the beads suspending after the reaction is settled on a bottom face and excitation light is cast onto the beads held settled on the bottom face, and then fluorescence or luminance caused is measured. By this method, a passage of the excitation light is not obstructed by the beads, and light emitted from the fluorescent or luminous substance reaches a detector directly so that a high detection sensitivity can be attained.
