摘要： Chromatography systems and methods for using carbon dioxide to separate one or more cannabis-derived compounds from other components of a mixture are generally described. Some of the methods described herein comprise transporting a mixture comprising a first cannabis-derived compound and one or more other components through a chromatography column containing a stationary phase comprising a packing material. In some embodiments, the mixture is transported through the column within a mobile phase that comprises carbon dioxide (e.g., supercritical CO 2 , liquid CO 2 ). The mobile phase may be substantially free of a co-solvent that is in liquid phase at standard room temperature and pressure. In some embodiments, the mobile phase is free of any co-solvent and comprises 100 vol% carbon dioxide. The first cannabis- derived compound may interact with the stationary phase and/or the mobile phase to a different degree than the one or more other components of the mixture, causing at least partial separation of the first cannabis-derived compound from the one or more other components within the column. Due to this separation, at least one fraction of the mobile phase that comprises the first cannabis-derived compound and is substantially free of the one or more other components of the mixture may be collected.

摘要： The present disclosure relates to methodologies, systems and apparatus for controlling pressure in a CO 2 -based chromatography system. A first CO 2 pump operates in constant flow mode and delivers CO 2 to a chromatography column, and liquid modifier is introduced to the chromatography column according to a gradient. A second CO 2 pump is disposed downstream of the column and operates in constant pressure mode to introduce CO 2 into a flow stream at an output of the column. Liquid modifier is also introduced into the flow stream at the output of the column according to a reverse gradient compared to the gradient entering the chromatography column.

摘要： Provided is a chiller and system that may be utilized in a supercritical fluid chromatography method, wherein a non-polar solvent may replace a portion or all of a polar solvent for the purpose of separating or extracting desired sample molecules from a combined sample/solvent stream. The system may reduce the amount of polar solvent necessary for chromatographic separation and/or extraction of desired samples. The system may incorporate a supercritical fluid chiller, a supercritical fluid pressure-equalizing vessel and. a supercritical fluid cyclonic separator. The supercritical fluid chiller allows for efficient and consistent pumping of liquid-phase gases employing off-the-shelf HPLC pumps. The pressure equalizing vessel allows the use of off the shelf HPLC column cartridges. The system may further incorporate the use of one or more disposable cartridges containing silica gel or other suitable medium. The system may also utilize an open loop cooling circuit using fluids with a positive Joule-Thompson coefficient.

In a fluid chromatography system, the mobile phase may include a pressurized mixture including a gas, such as C02, and prior to fraction collection and/or detection a portion of the gas is extracted via a gas/liquid separator. Cooling the substantially liquid phase eluent exiting the gas/liquid separator increases the solubility of any remnant gas still dissolved within the eluent. The increased solubility of the remnant gas prevents out gassing, provides more consistent flow of the eluent through the system, and results in more efficient collection and analysis of a desired sample.

The apparatus also includes computer software and hardware to control distribution of fluid through the apparatus, including switching between a multi-channel mode or a single channel ode; 2) analyze data collected by the one or more detectors; and 3) optimize separation of analytes by controlling solvent combinations, concentration gradients, pressure and temperature. The apparatus excludes additional backpressure regulators or pumps on individual channels. Also provided is a method of screening a sample, using supercritical chromatography, using the above apparatus, where multiple samples can be screened simultaneously with parallel processing.
