In a method of this type relating to selective plane illumination microscopy (SPIM), a reference structure is applied to the illumination light, at least one fluorescent reference object is positioned next to or in the sample, images of the reference structure for the illumination light, for the reference objects or of a sample structure that is suitable for use as a reference structure are captured from at least one detection direction and evaluated, the light layer is brought into an optimal position using the result, image information for the reference object and for the sample that has been captured from several detection directions is stored, the image information stored for the reference object is used to obtain transformation operators and these form the basis of the reconstruction of the three-dimensional reproduction of the sample from the image information that has been stored for the sample.
