摘要： This disclosure provides systems and methods for molecular identification and polymer (e.g., nucleic acid) sequencing using nanopores. The polymer may be passed through or in proximity to the nanopore and various subunits of the polymer may affect the current flowing through the nanopore. The various subunits may be identified by measuring the current at a plurality of voltages applied across the nanopore and/or membrane. In some cases, the polymerization of tagged nucleotides presents tag molecules to the nanopore that can be identified by measuring the current at a plurality of voltages applied across the nanopore and/or membrane. Also provided herein are systems and methods for sequencing both the sense and anti-sense strand of a double stranded nucleic acid molecule with a nanopore and methods for using ribonucleic acid (RNA) speed bump molecules to slow the passage of a nucleic acid molecule through or in proximity to a nanopore.

The disclosure also provides methods for sequencing nucleic acids using the disdosed tagged nudeotides. Provided herein are nudeotides with attached tags and methods for attaching tags to nudeotides. The tags can be attached by chemical reactions, such as "dick chemistry". In an aspect, the present disdosure provides a tagged nudeotide, comprising: (a) a poly-phosphate moiety having a terminal phosphate; and (b) a tag covalently coupled to the terminal phosphate of the nudeotide by a triazole, a 1,2-diazine, a disulfide, a secondary amine, a hydrazone, a thio-acetamide, or a maleimide-thioadduct.

Nucleic acid molecules can be sequenced with a high accuracy (e.g., greater than 97% in a single pass) using a chip comprising an array of independently addressable nanopore sensors at a density of at least about 500 sites per 1 mm. An individual nanopore sensor can include a nanopore in a membrane that is adjacent or in proximity to a sensing electrode.

摘要： This disclosure provides chips, systems and methods for sequencing a nucleic acid sample. Tagged nucleotides are provided into a reaction chamber comprising a nanopore in a membrane. An individual tagged nucleotide of the tagged nucleotides can contain a tag coupled to a nucleotide, which tag is detectable with the aid of the nanopore. Next, an individual tagged nucleotide of the tagged nucleotides can be incorporated into a growing strand complementary to a single stranded nucleic acid molecule derived from the nucleic acid sample. With the aid of the nanopore, a tag associated with the individual tagged nucleotide can be detected upon incorporation of the individual tagged nucleotide. The tag can be detected with the aid of the nanopore when the tag is released from the nucleotide.

摘要： Disclosed is an apparatus and corresponding method to capture individual representational motions made by a person while performing a neuropsychological test. A user of the apparatus and method may be allowed to view, edit, classify, and retrieve any captured and recorded motions made by the person. In one embodiment, a capture unit, such as a digitizer, captures the motions made by a person while performing a Clock Drawing Test, and the captured data may then be reported in a myriad of ways. The apparatus and method also provide a means to standardize administration and scoring of writing utensil and paper based neuropsychological testing administered to a person or control and test groups for domestic or international applications. By analyzing individual representational motions made by a person while performing a neuropsychological test, unexpectedly more information may be gleaned from the test than was available to physicians from only a completed drawing.

One method comprises receiving data from a digital device, the data comprising a plurality of digital device positions and a plurality of timestamps, each timestamp in the plurality of timestamps being associated with a digital device position in the plurality of digital device positions. The method further comprises determining a plurality of frequencies of hand movements of the subject based on the plurality of digital device positions and plurality of timestamps. The method further comprises determining a subportion of the data corresponding to frequencies of hand movements above a low tremor threshold, and determining a magnitude of tremors of the subject's hand based, at least in part, on the subportion of the data.
