摘要： The invention provides a method of determining activity of a protease. The method can include the steps of (a) providing a protease substrate including a protein moiety attached to a nucleic acid moiety and a ligand moiety; (b) contacting the protease substrate with a protease under conditions wherein the protease catalyzes cleavage of the protein moiety, thereby producing a proteolytic product wherein the nucleic acid moiety is separated from at least a portion of the protein moiety and the ligand moiety; (c) contacting the proteolytic product with a receptor under conditions wherein the ligand moiety binds to the receptor to form a complex; (d) separating the complex from the nucleic acid moiety, thereby forming a separation product including the nucleic acid moiety; (e) contacting the separation product with a probe nucleic acid under conditions wherein the nucleic acid moiety hybridizes to a complementary sequence of the probe; and (f) detecting hybridization of the separation product to the probe, thereby determining activity of the protease.

摘要： The invention provides systems and methods for evaluating synthetic reactions such as polymer synthesis reactions. Evaluation of a synthetic reaction product can be based on a quantitative measure of synthesis such as coupling efficiency or yield, and various parameters determined from a separation record obtained for the synthetic reaction product. The invention further provides methods for evaluating a separation method based on various parameters determined from a separation record. The invention can be used to select an appropriate treatment for a synthetic reaction product such as a separation treatment or treatments related to further processing for the desired product. The methods are particularly useful for automated selection of treatments for synthetic reaction products.

摘要： The invention provides a method of distinguishing small RNA from mRNA by contacting a biological isolate with a phosphate reactive reagent having a label moiety under conditions wherein the label moiety is preferentially added to the 5' phosphate of small RNA over the 5' cap structure of mRNA and distinguishing the small RNA from the mRNA according to the presence of the label. The invention further provides a method of identifying a plurality of different small RNAs by adding a unique extension sequences to different small RNA sequences and identifying the extended small RNA sequences. Furthermore, the invention provides diagnostic methods for determining presence of a disease or condition such as cancer. Also provided are prognostic methods for determining progression of a disease or condition or for monitoring effectiveness of a treatment for a disease or condition.
