    • Methods, processes and systems for procuring, isolating and cryopreserving at least one viable, multipotent maternal placental stem cell is provided. Viable maternal placental stem cells are also provided. The maternal placental stem cell of the invention expresses the cell surface marker CDl 17 and at least one of the cell surface markers selected from the group consisting of CD29, CD44, CD73, CD90, CD105, CD166, SSEA-3 and SSEA-4 and has low or no expression of at least one of the cell surface markers selected from the group consisting of CD34, CD45, CD133, TRA-1-60 and TRA-1-81. The methods and process comprise generally obtaining a piece of placental tissue from a whole placenta, disaggregating the placental tissue with mechanical separation or enzymatic digestion, collecting and concentrating placental cells comprising maternal placental cells with centrifugation and optionally cryopreserving the placental cells. Additional steps for selecting and culturing the maternal placental stem cells is provided.

    • An apparatus for preparing an elongate fluid-containing member (10) such as an umbilical cord comprises a housing (14) and support elements (30) in the housing (14) for maintaining the umbilical cord (12) in substantially linear configuration. The support elements (30) are generally horizontally oriented and are pivotally mounted to the housing. A clamping device (42) in the housing (14) applies at least four clamps or staples (50) to the umbilical cord (12) while a severing device (58) divides the umbilical cord (12) to form at least two separate sections (114). A transfer mechanism (80) in the housing (14) automatically transfers the severed umbilical cord sections (114) to respective receptacles (74), the transfer mechanism (80) including a device operatively connected to the support elements (30) for tilting those elements, the transfer mechanism (80) further including guide funnels (36) disposed in the housing (14) below the support elements (30) for guiding the severed sections of the umbilical cord (114) into the receptacles (74). A closure assembly (150) is provided for automatically closing the receptacles.

The methods involve seeding a sufficient amount of cord blood cells with a sufficient amount of menstrual cells under co-culture conditions suitable to promote expansion of the cord blood cells, and co-culturing the cord blood cells with the menstrual cells under culture conditions that support at least two or more population doublings of the cord blood cells.
