摘要： A novel class of cationic phospholipids and a novel method for their synthesis are disclosed. The class of phospholipids comprises phosphotriester derivatives of phosphoglycerides and sphingolipids. It has been unexpectedly found that liposomes comprising one or more of these cationic phospholipids are effective in the lipofection of nucleic lacids. These novel phospholipids are particularly attractive because they are extremely cheap and easy to make.

The method comprises the following steps: A. dissolving the fat concentrate in a solvent mixture of an essentially polar organic solvent and an essentially non-polar organic solvent, B. withdrawing a phase consisting mainly of the non-polar organic solvent and phospholipids dissolved therein, C. adding to the phase withdrawn in step B an organic solvent of intermediate polarity at a temperature of about 13.degree.-25.degree. C., thereby forming a precipitate comprising mainly sphingomyelin, together with a viscous phase and a solvent phase, and then D. withdrawing the precipitate and the viscous phase, and separating them from one another.

摘要： Compositions and methods for use in generating fast ratiometric voltage-sensitive fluorescence changes in single or multiple cells systems. A first reagent is a membrane-bound hydrophobic fluorescent anion which rapidly redistributes from one face of the plasma membrane to the other in response to the transmembrane potential, as described by the Nernst equation. A voltage-sensitive fluorescent readout is created by labeling the intracellular or extracellular surface of the cell with a second reagent comprising a fluorophore which can undergo energy transfer with the first reagent or a quencher for the first reagent. Quenching or FRET between the two reagents is disrupted when the membrane potential is depolarized, because the anionic first reagent is pulled to the intracellular surface of the plasma membrane far from the asymmetrically bound second reagent. In preferred embodiments of the invention, the first and second reagents are bound together by a suitable linker group.

摘要： The present invention provides materials which comprise a surface having pendant zwitterionic groups Z comprising, as anion, a phosphate, sulphonate, carboxylate or phosphate-ester group or a phosphate ester group in which one or more of the ester oxygen atoms is replaced by --S--, --NH-- or by a valence bond and, as cation, an ammonium, phosphonium or sulphonium moiety, provided that where the anion is a phosphate-ester group or derivative thereof or sulphonate and the cation is a trialkyl quaternary ammonium group (the alkyl groups each containing from 1 to 4 carbon atoms) and the group linking the anion and cation moieties is an alkylene group, the alkylene group, contains at least 5, preferably at least 6, and more preferably at least 7, carbon atoms. The materials have improved biocompatibility for instance as evidenced by reduced protein deposition and reduced platelet activation.
