摘要： This invention demonstrates that the PEG10 protein suppresses apoptosis and promotes cancer cell growth. Furthermore, PEG10 protein was found to be degraded through interaction with SIAH1 protein. This invention provides methods of regulating cell growth and cell death using the PEG10 protein. Furthermore, the present invention provides methods of screening for novel anticancer agents which use the PEG10 protein. The screening of this invention enables development of pharmaceutical agents that induce apoptosis and suppress cell growth by specifically targeting cancer cells. In particular, since the PEG10 protein is specifically expressed in hepatoma cells, it is an ideal target molecule for diagnosis and treatment of hepatoma.

For example, the methods disclosed herein can reliably detect very early-stage colorectal cancers. In one embodiment, the diagnostic method involves the scoring of gene expression profiles that discriminate between adenomas and carcinomas. The profile score calculated acts as diagnostic indicator that can objectively indicate whether a sample tissue is non-cancerous, pre-cancerous, or cancerous. The present invention further provides methods of diagnosing colorectal tumors in a subject, methods of screening for therapeutic agents useful in the treatment of colorectal tumors, methods of treating colorectal tumors and method of vaccinating a subject against colorectal tumors.

A clone having a novel sequence including a sequence homologous to that of the transcriptional control protein of a yeast was selected from among the above clones and its whole nucleotide structure was determined. It was confirmed that the protein encoded by the gene of the clone was a novel human transcriptional control protein (RPDL protein). Further, an expression vector for expressing the protein and a transformant obtained by transforming with such an expression vector can also be prepared.
