    • 본발명은크로마토그래피컬럼을작동시키는방법에관한것이다. 이방법은컬럼충전물(column packing)을포함하는크로마토그래피컬럼의작동동안 2개이상의간격으로적어도하나의이동상전이전선(mobile phase transition front)의컬럼출구신호및 누적유량파라미터를수집하는단계를포함한다. 적어도하나의이동상전이전선에대해수집된컬럼출구신호및 누적유량파라미터에기초하여모델감마누적분포곡선이결정된다. 모델감마누적분포곡선의파라미터를사용하여적어도하나의이동상전이전선에대해이론단상당높이(HETP) 값이계산되고, 계산된 HETP 값에기초하여크로마토그래피컬럼충전물의품질이평가된다. 일상적인컬럼모니터링동안, HETP에서의불리한경향이관찰되거나관리한계선이초과되는경우에는, 확인된배치(batch)에대한생성물품질을보장하기위해용리생성물품질, 컬럼공정성능, 및/또는불순물제거데이터가평가되어야한다. 생성물품질또는컬럼성능중 임의의것이기준세트에합격하지못한다면, 적절한교정활동, 예컨대컬럼의컨디셔닝, 재충전(repacking) 또는교체, 그리고적격성평가(qualification)가추가사용을위한방출전에수행되어야한다.

An ultrahigh-pressure cation exchanger dual on-line solid phase extraction/capillary reverse-phase liquid chromatography system comprises a sample inlet valve(100) including an inlet port(101) of strong cation exchange solvent, a sample inlet port(102), a sample outlet port(103), sample storage loop connection ports(104,104') and a column selection valve connection port(105); a column selection valve(200) which determines whether the inlet sample is transferred to the first column valve or to the second column valve, and includes a first column valve connection port(202) and a second column valve connection port(203); a first column valve(300) which is communicated with a first solid phase extraction column(302) and a first reverse-phase liquid chromatography column(303); a second column valve(400) which is communicated with a second solid phase extraction column(402) and a second reverse-phase liquid chromatography column(403); a solvent selection valve(500); a second solvent inlet valve(600); a second solvent supply pump; a first solvent supply pump; and a strong cation exchange solvent supply pump.

CONSTITUTION: An inverse gas chromatography data processing method includes the following: objective materials are introduced into a hollow column(S11); the column is mounted in a gas chromatography device(S12); the analyzing condition of the gas chromatography device is set(S13); first chromatogram is obtained by introducing a first mole number of a searching material into the gas chromatography device using carrier gas(S14); second chromatogram is obtained by introducing a second mole number of the searching material into the chromatography device, and the second mole number is different from the first mole number(S15); absorption free energy is respectively obtained from the first chromatogram and the second chromatogram; and final absorption free energy without a searching material is extrapolated from the absorption free energy(S16).
