A production method of interleukin-18 comprises the steps of: (a) inserting a heterogenous gene into a vector comprising GPD promoter or AG promoter, extracellular secretion signal sequence, cytokine gene and transcription termination signal sequence to prepare a recombinant vector; (b) transforming yeast with the recombinant vector; and (c) culturing the transformed yeast in a medium.
