CONSTITUTION: A vector comprises (a) GPD(glyceraldehyde-3-phosphate dehydrogenase) promoter(GenBank accession number M13807) or AG promoter(GenBank accession number J01314); (b) extracellular secretion signal sequence; (c) cytokine gene; and (d) transcription termination signal sequence, wherein the extracellular secretion signal sequence is amylase 1A secretion signal sequence (Ramy1A, GenBank accession number X16509); the cytokine gene is a gene encoding interleukin-18 mature protein; the vector is pYEGIL18 or pYEAGIL18. A production method of interleukin-18 comprises the steps of: (a) inserting a heterogenous gene into a vector comprising GPD promoter or AG promoter, extracellular secretion signal sequence, cytokine gene and transcription termination signal sequence to prepare a recombinant vector; (b) transforming yeast with the recombinant vector; and (c) culturing the transformed yeast in a medium.
