    • PURPOSE: An encapsulated cell indicator system is provided, thereby determining the differentiation state of cells during the culture thereof in real time and in a non-destructive manner. CONSTITUTION: An encapsulated cell indicator system comprises (a) indicator cells having a signal-responsive element operably linked to a reporter gene; (b) encapsulating material; and (c) a permeable membrane, wherein the indicator cells are encapsulated in the encapsulated material; the encapsulated material and the indicator cells are surrounded by the permeable membrane; the indicator cells comprise embryonic stem cells or bone marrow stem cells; the indicator cells comprise human cells; the indicator cells comprise mouse cells or pig cells; the reporter gene encodes beta-galactosidase, green fluorescent protein or luciferase; the encapsulating material comprises alginate, collagen, gelatin, chitosan, polylactic acid, polyglycolic acid, or polylactide/glycolide copolymer; and the permeable membrane comprises polyethylene terephthalate membrane, nylon mesh, porous nylon membrane, or porous polytetrafluoroethylene(PTFE/Teflon).

    • Methods for producing cells for transplantation are provided to enhance cell engraftment and cell survival rate of stem cells, monitor differentiation induction of stem cells in vitro, and collect stem cells for transplantation. The method for producing cells for transplantation comprises the steps of: (a) supplying immortalized bone marrow stem cells; (b) culturing the immortalized bone marrow stem cells under conditions that induces the differentiation of the immortalized bone marrow stem cells into vascular smooth muscle cells, endothelial cells, epicardial cells, adipocytes, osteoclasts, osteoblasts, cardiac fibroblasts, macrophages, neuronal progenitors, neurons, astrocytes, skeletal muscle cells, smooth muscle cells, pancreatic precursor cells, pancreatic beta-cells or hepatocytes; (c) monitoring the differentiation state of the cells of step(b); and (d) collecting the cells of step(b) when 10-100% of cells express a detectable amount of proteins specific to the cell type, wherein the method further comprises the step of transplanting the cells into mammals.

    • PURPOSE: A method for preparing cell for cell transplantation is provided, thereby preparing cells for transplantation to mammal heart tissue in higher yield, so that it can treat a disorder by unstable heart function. CONSTITUTION: The method for preparing cell for cell transplantation comprises the steps of: (a) supplying immortalized bone marrow stem cell; (b) culturing the immortalized bone marrow stem cell in a medium containing IGF-1 under condition to induce differentiation of immortalized bone marrow stem cell into myocardial cytoblast; (c) monitoring the differentiation status of immortalized bone marrow stem cell; and (d) collecting the immortalized bone marrow stem cell when 50% of more of the cells are myocardial cytoblast, wherein the immortalized bone marrow stem cell is derived from a mammal to be transplanted; the mammal is human; and the concentration of IGF-1 is 0.1 to 25 ng/ml.
