    • PROBLEM TO BE SOLVED: To provide an efficient directed differentiation method of hESC (human Embryonic Stem Cell or human Embryonic Germ Cell) as a starting material for generating a useful cell in a cell therapy application.SOLUTION: There is disclosed a method for identifying a differentiation factor which can promote the differentiation of human definitive endoderm cells in a cell population comprising human cells, and the method comprising the step of obtaining a cell population comprising human definitive endoderm cells, the step of supplying a candidate differentiation factor to the cell population, the step of measuring expression of a marker in the cell population at the first time point, the step of measuring expression of the same marker in the cell population at the second time point, and the step of determining whether expression of the marker in the cell population at the second time point is altered as compared to the expression of the marker in the cell population at the first time point, wherein the second time point is subsequent to supplying the cell population with the candidate differentiation factor, and the alteration in expression of the marker in the cell population indicates that the candidate differentiation factor can promote the differentiation of the human definitive endoderm cells.
