    • PURPOSE:To provide a protein originated from human psoriasis scales, having a specific mol. wt. and an amino acid sequence, inhibiting the activity of cathepsin L which is a cysteine protease originated from an animal, and useful for preventing and treating osteoporosis, etc. CONSTITUTION:This new psoriasis scale-originated protein having a mol. wt. of approximately 43 kDa by a SDS-PAGE method, having amino acid sequences of formulas I-V, etc., in the molecule, inhibiting the activity of cathepsin L which is a cysteine protease originated from an animal but not inhibiting the activities of cathepsin B and cathepsin H is obtained by treating scales collected from a psoriasis patient in a buffer solution with a homogenizer for uniformly extracting the scales, centrifuging the extract, collecting the supernatant, concentrating the supernatant with an ultrafiltration membrane, subjecting the concentrated solution to a gel-filtration treatment for obtaining a fraction inhibiting papain and having a mol.wt. of approximately 40000, allowing the fraction to pass through an anion exchange column, applying the passed fraction to a cation exchange column, fractionating the treated fraction by a NaCl gradient method, and subsequently purifying the active fraction by an isoelectric point chromatography.
