摘要： PROBLEM TO BE SOLVED: To provide a method for monitoring conditions of a food product, especially the degree of heat treatment of the food product, the degree of sterilizing treatment of the food product or freshness of the food product by using an enzyme activity in the food product as an index.
SOLUTION: 1. The method for monitoring the conditions of the food product comprises the following two steps. (1) a step of bringing the food product or its extract into contact with β-galactosidase as a substrate of (first step) and (2) a step of detecting a reaction product produced by the first step and thereby assaying the β-galactosidase activity in the food product (second step). 2. The method for monitoring the conditions of the food product comprises the following two steps. (3) a step of bringing the food product or its extract into contact with a luciferin galactoside, luciferase, adenosine 5'-triphosphae (ATP) and a metal ion (first step) and (4) a step of detecting the light emission produced by the first step and thereby assaying the β-galactosidase activity in the food product (second step). 3. A luciferin derivative, the luciferase, ATP and the metal ion are contained in a reagent for monitoring the conditions of the food product.

摘要： PURPOSE:To obtain a new sorbitol dehydrogenase produced by the culture of microbial cell belonging to the genus Pseudomonas and capable of producing sorbitol dehydrogenase, extremely useful for the enzymatic assay of D-sorbitol and usable e.g. for the diagnosis of diabetes. CONSTITUTION:This new sorbitol dehydrogenase useful e.g. for the enzymatic assay of D-sorbitol is produced by culturing a microbial strain belonging to the genus Pseudomonas and capable of producing sorbitol dehydrogenase [e.g. Pseudomonas sp.KS-E1806 (FERM P-14229)] in a medium. The enzyme is effective to oxidize D-sorbitol in the presence o NAD to produce D-fructose and NADH and specifically act on D-sorbitol and galactitol, has an optimum pH of about 10 and stable pH of 5.5-10.5 (treatment at 30 deg.C for 4hr), completely deactivated at pH =11.0, has optimum working temperature range of about 50 deg.C in 100mM tris-HCl buffer solution (pH 9.0), is strongly inhibited with HgCl2 and has a molecular weight of about 64,500+ or -5,000 (gel- filtration).

摘要： PROBLEM TO BE SOLVED: To quickly quantify the level of minute amounts of D-sorbitol or galactitol or their mixture in a sample such as of blood, tissue, body fluid, food or the like in a simple operation and in high sensitivity. SOLUTION: First, a dehydrogenase reactive to D-sorbitol and/or galactitol in the presence of NAD is added to a sample to carry out a reaction. Secondly, the resultant NADH is phosphorylated by the use of NADH kinase to form NADPH. Subsequently, cycling reactions are conducted by using an enzyme or electron carrier capable of oxidizing the NADPH into NADP and that capable of reducing NADP into NADPH, and the change in substrate consumed or product formed in the cycling reactions is detected, thus quantifying the D-sorbitol and/or galactitol in high sensitivity.

The enzyme is obtained from a cultured product of a strain belonging to the genus Bacillus and having the ability to produce the α-glucosidase.
