摘要： PURPOSE:To obtain a genetically stable biological preparation accompanied with extremely little danger of biohazard because of avirulence by using an attenuated Marek's disease virus genomic DNA as a virus vector. CONSTITUTION:As a genetically stable attenuated virus free from reversion to a virulent strain is used, the abovementioned method facilitates mass- production of a biological preparation safe considering from the viewpoint of biohazard during the production process at a low cost. The abovementioned method also realizes a method for production of a recombinant gene using a virus vector, producing a product safe and effective as an active component of a live vaccine, etc., and having merits such as low cost and reduction of labor in use and practical application of the preparation. The realized achievement is due to excellent usefulness as a virus vector for each of an attenuated Marek's disease virus, HVT, O1 strain and MDV.C2 strain.

摘要： NEW MATERIAL:A surface antigen of hepatitis B virus, expressed by the formula (HBs antigen). USE:A diagnosticum or a vaccine for hepatitis B virus infection. PREPARATION:For example, a serum of a patient infected by hepatitis B virus (HBV) is centrifugated at 10,000rpm to separate a supernatant liquid. This supernatant liquid is further centrifugated at 28,000rpm for 4hr and HBV virus particles are obtained from the sediment. The virus particles are treated with a protease, etc., and further treated with a water saturated phenol to obtain DNAs. Then the DNAs are treated with restriction endonucleases, subjected to cloning and then combined with a replicable expression vector to form a recombinant DNA. The recombinant DNA is then introduced into a microorganism to transform, and this transformant is cultured to obtain the compound expressed by the formula from the culture fluid.

A clone S22 having a length of 2,500bp, containing 5'-terminal of virus RNA and containing a gene region coding V3 protein of Japanese encephalitis is separated from the cloned product. XboI linkers are linked to both terminals of the clone to obtain a clone S22X. A plasmid pJM105 for the manifestation of V3 protein gene is constructed by using a plasmid pS22X and a manifestation vector YEp133PCT.

These virus particles are obtained by selecting a clone containing a cDNA encoding the objective antigen from a cDNA group prepared from a non-A non-B type hepatitis virus genome RNA fragment, cutting out the cDNA, linking the cDNA to a manifestation vector composed of a viral gene, culturing the prepared recombinant virus and manifesting.

摘要： PURPOSE:To provide a mutant of Bordetella pertussis phase-I, free from pertussis toxicity and producing a pertussis toxin protein devoid of a part of subunit capable of deriving an antibody neutralizing the biological activity of pertussis toxin. CONSTITUTION:Phase-I cell of Bordetella pertussis is cultured by conventional method and washed under centrifugal treatment. The obtained bacterial cells are suspended in a tris-maleic acid buffer solution at a concentration of 10 mol./ml, added with 25-50mug/ml of nitrosoguanidine as a mutagenic agent and shaken for 60min. The product is subjected to centrifugal washing in a liquid medium, properly diluted and cultured on a solid medium by plate culture. A colony on the plate is picked up, inoculated in a liquid medium, cultured by shaking culture and screened with respect to the supernatant liquid of the cultured product to obtain a 79G strain (FERM BP-1902) which is inert to the clustering of CHO cell and reactive with a polyclonal antibody of pertussis toxin. The 79G strain is cultured in a medium added with a cyclodextrin and a pertussis toxin or protein devoid of a part of subunit, especially subunit S1 is separated from the cultured product.

摘要： PURPOSE:To obtain a high-purity pertussis toxin (PT) and fibrous hemagglutinin (F-HA) in high yield at an extremely low cost, by cultivating Bordetella pertussis in a culture medium containing cellulose (derivative). CONSTITUTION:Cellulose or a derivative thereof or both are added and blended with a basal culture medium (e.g. Cohen.Wheeler culture medium) used for cultivating Bordetella pertussis so as to provide 0.01-2.0wt.% final concentration and the Bordetella pertussis is cultivated in the resultant modified culture medium at 30-37 deg.C for 20-80hr. The obtained culture is subsequently separated into a cultivation supernatant and microbial cells. The above-mentioned cultivation supernatant is then purified to recover PT and F-HA. Formalin is subsequently added and mixed to detoxify the purified PT and F-HA fraction and formalin is removed by dialysis. The resultant dialyzate is diluted with a phosphoric acid buffer solution, etc., to provide a vaccine concentrate, which is then used to prepare a mixed vaccine. Gelatin (derivative) in an amount of 0.1-5.0wt.% as a stabilizer is added to remarkably improve heat and cold resistance.

摘要： PURPOSE:To remove allergen originated from a transformant and purify a gene manifestation substance, by using a series of purification steps comprising a combination of adsorption and desorption with silicic acid, treatment with activated carbon, density gradient centrifuge and equilibrium density centrifuge. CONSTITUTION:An extract of a primary product or secondary product of gene manifestation is added with about 0.1-10w/v% silicic acid and stirred at 10-30 deg.C for 10-100min to effect the adsorption of gene manifestation product. The adsorbed substance is eluted with a proper eluent and the obtained solution is added with 0.1-10w/v% activated carbon and stirred at 10-30 deg.C for 5-30min to obtain a supernatant liquid free from impurities. The supernatant liquid is adjusted to 7-9pH, concentrated and subjected at least twice to a source density gradient centrifuge (about 10-20w/w%-40-50w/w%, 20,000-40,000rpm, about 2-30hr). A fraction containing said product is subjected at least twice to potassium bromide equilibrium density gradient centrifuge (about 10-50%, 30,000-60,000rpm, about 30-80hr) to obtain a purified gene manifestation product.
