摘要： PROBLEM TO BE SOLVED: To discriminate difference between prostatic cancer and prostatic hypertrophy within a region where an amt. of tPSA(prostate specific antigen) is 3-10 ng/ml, by directly measuring a ratio of fPSA to tPSA without independently measuring fPSA and tPSA. SOLUTION: A specimen is brought into contact with a solid phase wherein a first receptor having compatibility to both of a free prostate specific antigen, a prostate specific antigen forming a composite is fixed on a carrier to selectively collect a definite amt. of the prostate specific antigen in the specimen to separate the solid phase and the specimen, and a labelled second receptor having compatibility to the free prostate specific antigen is brought into contact with the separated solid phase. By this constitution, a labelling substance is bonded to the free prostate specific antigen contained in the whole amt. of the prostate specific antigen collected by the solid phase, and the solid phase and liquid phase of this reaction mixture are separated to measure the labelled substance amt. of the separated solid phase or the liquid phase.

摘要： PURPOSE:To simply measure isolated type thyroxin in blood, by labelling a bond of thyroxin and polypeptide with I or the like to perform a radiation immunoassay using the labelled antigen obtained. CONSTITUTION:A T4-polypeptide bond is obtained from thyroxin (T4) and polypeptide by a glutaric aldehyde method. The bond thus obtained is inactive to a thyroxin-bonded globulin (TBG) and albumin but makes a bond reaction with T4. Then, the bond is labelled with, for example, I or the like to make a labelled antigen, which is used to measure thyroxin T4 in blood by a radiation immunoassay method. Here, for example, a bond of thyroxin and sex gland stimulating hormone labelled with I is used. Thus, the use of the labelled antigen enables sensitive measurement of isolated type thyroxin without being affected by coexisting components in blood, TBG and the like this method is highly effective for the decision on the function of a thyroid gland.

摘要： PURPOSE:To simply measure isolated type thyroxin in blood, by performing an enzyme immunoassay using a bond of thyroxin and beta-D-galactosidase as enzyme as labelled antigen. CONSTITUTION:A T4-beta-D-galactosidase is obtained by maleimide method from thyroxin (T4) and beta-D-galactosidase as large molecular weight protein. The bond thus obtained is inactive to thyroxin-bonded globuline (TBG) and albumin but makes a bond reaction with T4. The T4-protein bond is used as labelled antigen to measure thyroxin T4 in blood by an enzyme immunoassay. Here, the bond can be used as enzyme labelled antigen as T4 is coupled to large molecular weight enzyme. Thus, the use of the labelled antigen enables sensitive measurement of isolated type thyroxin without being affected by coexisting components in blood, TBG and the like and this method is highly effective for the decision on the function of a thyroid gland.
