    • PURPOSE:To enable the highly accurate measurement of an antigen in a short time by applying force to an obtained sediment in another direction different from the sedimentation direction upon applying centrifugal force, followed by measuring its dimensional change. CONSTITUTION:A specimen, in which the antigen to be measured is likely to be contained, is reacted with a carrier, in which the corresponding antibody is contained in a solid phase, directly or dilutedly. An antigen-antibody reaction can be performed under a normal condition. The resulted reactant is forcibly sedimented by a centrifugal operation to form a sediment of a reactant. The condition for the centrifugal operation is not preferably restricted as far as the reactant can be sedimented, for example about ten minutes at 1200 rpm. Next, an external force is applied to the obtained sediment in another direction different from the sedimentation direction upon applying the centrifugal force, followed by measuring the dimensional change of the sediment. Although the sediment maintains the pellet-like shape if an antigen-antibody reaction has taken place, it can be observed that the carrier particle flows in a tear-like fashion if no antigen-antibody reaction has taken place.

    • PROBLEM TO BE SOLVED: To measure a low concentration of antigen in a sample with high sensitivity by bringing about a reaction between an antigen present in a sample solution and an antibody carried by magnetic particles, gathering the magnetic particles, suspending them in a small amount of liquid, mixing carried non-magnetic insoluble particles into reaction, and measuring the degree of agglutination of the obtained reaction mixture. SOLUTION: An antigen to be an object to be measured is a microorganism or an antigen derived from a microorganism and is tubercle bacillus, staphylococcus, etc. There arc organic high-molecular matter and inorganic micro powder containing magnetic bodies as magnetic particles. There are organic high-molecular matter and inorganic micro powder as non-magnetic insoluble particles. In measurement, a sample solution and a magnetic particle liquid are mixed to bring about a reaction between an antigen present in the sample solution and an antibody carried by magnetic particles. After achieving equilibrium in the reaction. a magnetic field is applied to gather not-yet-reacted magnetic particles and carried magnetic particles. The gathered magnetic particles are suspended in a small amount of liquid to concentrate the antigen selectively. After the non-magnetic insoluble particle liquid carrying the antibody is mixed with the suspension to bring about a reaction, the degree of agglutination is measured.
