摘要： PURPOSE:To determine the antibody to the antigen A in human serum, by using a conjugate of normal human immunoglobulin and an antibody derived from an animal to the antigen A as a standard substance. CONSTITUTION:In order to measure the anti-thyroxine antibody in human serum, an enzyme labelled thyroxine is prepared as a standard antigen (a), a 10mM phosphate buffer solution containing 0.5% gelatin, 0.1% bovine serum albumin and 0.3M NaCl as a buffer solution (b), a 0.1ml polystyrene column packed with anti-human IgG antibody insolubilizing cephalose as a column (c), 33mM o-nitrophenyl-beta-D-galactoside solution as a substrate (d) and a 80mM sodium carbonate solution as a color former (e). 500mul of the labelled antigen (a) is added to 100mul of a diluted standard solution and 100mul of diluted serum to perform reaction for 2hr at 37 deg.C. After the column (c) is washed with the buffer solution (b), the substrate (d) is made to flow through the column (c) and, after the column (c) is allowed to stand for 90min at 25 deg.C, the absorbancy of the washing solution obtained by washing the column (c) with the color former (e) at 420nm is measured.

摘要： PURPOSE:To determine quantitatively many antigens with good sensitivity by using the same antibody insolubilizing carrier by using an antibody labeled by heterologous immune globulin, enzyme labeled antibody and the insoluble carrier insolubilized with the antibody for the heterologous immune globulin. CONSTITUTION:The antigen A, the antibody labeled with the heterologous immune globulin B and the antibody C labeled with the enzyme D are brought into reaction with each other, by which the immune complex B-C-A-C-D is obtd. Such immune globulin is conjugated with the carrier F insolubilized with the antibody E to the globulin B to obtain the conjugate D-C-A-C-B-E. The activity of the enzyme D in the conjugate is thereafter measured, by which the antigen A is quantitatively measured. The quantitative determination of the many antigens A with good sensitivity with the same antibody insolubilized carrier F-E is made possible by using the same globulin B according to the above-mentioned method.

A brain tumor-specific antigen corresponding to the monoclonal antibody specific to the glioma-specific antibody and labeled with a detectable substance and a factor capable of bonding to the monoclonal antibody specific to the glioma- specific antigen and insolubilized on a water-insoluble carrier are added thereto and the resultant mixture is reacted to detect the detectable substance bonded to the water-insoluble carrier.
